Purpose: This study established the model of exercise-induced renal ischemia-reperfusion in rats by high-intensity exercise,observed the changes of expression of SOCS-1,NF-?Bp65 and related inflammatory cytokines in different time courses,trying to provide scientific theory basis of the physiological mechanism of ischemia-reperfusion injury induced by exercise-induced renal injury and ways about how to prevent and postpone the occurrence and development of exercise-induced renal injury in the future.Methods: 8-week-old male wistar rats were randomly divided into quiet control group(C Group),middle-intensity exercise(T Group)and high-intensity exercise Group(LT group),8 rats in each group according to the different time points.Rats in C Group do not take any exercise.Rats in T Group carried on the 6 weeks moderate intensity swimming exercise.Rats in LT Group had the same exercise with T Group in first 3 weeks then had the increment load swimming exercise.We detected the NGAL,Scr,BUN concentration by Radioimmunoassay to see if we have success of exercise-induced renal ischemia-reperfusion model.At the last exercise,blood was sampled at the moment ending exercise,6 hours after exercise,24 hours after exercise in EDTA-anticoagulated tubes for the test,at the same time taking the rats' bilateral kidneys which were stored at-80? for analysis.We measure SOCS-1 and NF-?bp65 protein content of renal tissue by western-blot.Results:(1)The renal tubule epithelial cells edema and lumen dilatation were found in the LT group,but a little edema occurred in the T group.(2)The expression of SOCS-1 protein in T group and LT group was significantly lower than that in C group immediately after exercise and 24 hours after exercise(p<0.05).There was no significant difference between groups(3)The expression of socs-1 and NF-?bp65 in the kidney tissue of T group and LT group showed a tendency of decreasing first,then rising and then decreasing.Conclusion:(1)After 6 weeks of high-intensity exercise and middle-intensity exercise,the renal function of rats was disturbed to a certain extent.The expression of SOCS-1and NF-?Bp65 in kidney of convalescent stage decreased at first and then increased and then decreased,which played a certain regulatory role.(2)The renal dysfunction induced by 6 weeks high-intensity exercise was more serious than that of middle-intensity exercise,and the expression of NF-?Bp65 in renal tissue was significantly decreased 24 hours after exercise.
