Objective: through the single factor experiment of single factor extraction,use Box-Behnken software for statistical analysis,the experimental value is compared with the predictive value,establish the optimal extraction conditions of polysaccharide extracts of Maca,Maca polysaccharide in rats and explore its influence on rat anti fatigue,improve athletic ability and the analysis of its mechanism,in order to optimize the extraction process of Polysaccharide from Maca,and the development and utilization of health care products or as a supplement to provide experimental basis for movement.Research methods: the optimum extraction conditions of the factors determining the single factor experiment,statistical analysis was carried out by Desigin-Expert8.0.6 Box-Behnken,using multiple regression method,model test design of two multinomial fitting,the theory of optimum extraction scheme,the best extraction rate is measured to detect and determine the best extraction scheme and theoretical value;the experimental preparation of Maca as to the tested polysaccharide gavage animal experiments were carried out in SPF rats,80 male SD rats were randomly divided into 4 groups,negative control group,S Maca polysaccharide low dose group ML(130mg/Kg-bw/d),Maca polysaccharide in MM dose group(270mg/Kg-bw/d),Mary Maca polysaccharide high dose group MH(800mg/Kg-bw/d),gastric lavage solution was 0.1ml/10 g bw/d,after intragastric administration for 30 days respectively from each group were randomly selected 10 rats by force liver glycogen,only for the remaining 40 The exhaustive swimming time and the biochemical indexes of rats' exercise ability were measured.Research results: 1.Maca polysaccharide ultrasonic assisted extraction process optimization: the single factor experiment results: the optimal solid-liquid ratio was 1:20g/ml,the optimum ultrasonic time 30 min,ultrasonic temperature is 60 DEG C,Box-Behnken Desigin-Expert8.0.6 software for statistical analysis concluded that the extraction rate of regression equation: the extraction rate of =17.61+0.41*A-003*B+0.031*C-0.32*A*B+0.27*AC+0.014*B*C-0.16*A2-1.20* B2-1.26*C2 Theory: the best extracting method the ratio of material to liquid 1:22g/ml,ultrasonic time 28 min,ultrasonic temperature is 59 degrees centigrade,the extraction rate was 17.6754%.2.the effect of Maca polysaccharide on the exercise ability of rats: 1.There was no significant difference in weight between the first and fifth weeks of the rats(P > 0.05).Both middle and high dos es of Maca polysaccharide could significantly improve the swimming time of exhausted rats.3.The Maca polysaccharide group(ML,MM,MH)can effectively inhibit the growth of lactic acid in rats and accelerate the elimination of lactic acid.The effect of high dose MH group is the most obvious.4.The conte nt of BUN in swimming rats was reduced by different doses of Maca polysacc haride.Reduce the content of serum pyruvicacid in rats were low in the high Maca polysaccharide,high dose of Maca polysaccharide on reducing the content of MDA was the most significant.6.Different doses of Maca polysaccharide increased liver glycogen storage in rats,the high dose of Maca polysaccharid e to improve the effect of liver glycogen content of rats was the most significant.Research conclusion: 1.the optimum extraction conditions of Maca polysa ccharide are as follows: the ratio of material to liquid is 1:22g/ml,the ultrason ic time is 30 min,the ultrasonic temperature is 60,and the extraction rate is 17.6754%.2.Maca polysaccharide can enhance lactic acid clearance rate,improve the body's ability to recover;reduce the content of exhaustive swimming BUN,multi degree by slowprotein;improve the antioxidant capacity of the body,reduce the damage of free radicals on the cell membrane,protect the muscle fiber function to maintain normal muscle contractility;improve energy reserves;prolonged exhaustive swimming time;in summary,Maca can delay the appearance of fatigue,enhance endurance,enhance athletic ability,can be developed into sports supplements,health care products.
