Objective:The study established a diet induced obesity model in female clean SD rats,followed by aerobic exercise(swimming as an example)combined with the experimental intervention of controlling diet.On the one hand,observe the change of the 6 week experimental period of obese rats body weight,body length,abdominal circumference and Lee s index etc obesity index,explore the swimming training combined with diet intervention effect on obesity;on the other hand,using double antibody sandwich enzyme-linked immunosorbent assay and immunohistochemical analysis of the expression of the content of experimental method.And the hypothalamic arcuate nucleus positive cells in rats plasma and hypothalamic neuropeptide Y 6 weeks after the experiment,to provide new ideas for future formulation of scientific guidance and prevention of obesity weight loss and exercise prescription,and to provide theoretical basis for the future and reduce the onset of chronic diseases related to obesity,and then combined with the controlled expression of food on the central and outside week of NPY provides a new research direction for the further study of sports.Methods:After 130 days of adaptive feeding to 130 female clean grade SD rats,8 rabbits were randomly selected as normal control group(NC: n=8),free dietary common diet,and obesity models were established for the rest of the rats.After free and high-fat diets,48 obese rats were selected after eight weeks for adaptive swimming for three days.The obese rats were then randomly divided into obesity control group(OC: n=8)and diet group(D).: n=8),sports immediate group(E0: n=8),three-hour sports group(E3: n=8),sports dieting immediate group(ED0: n=8)and sports three-hour diet group(ED3:n = 8)Six groups,90 min/day,6 days/week of swimming training combined with dietary control of experimental interventions.Among them,the normal control group had normal diet and the obese control group had high-fat diet.The two groups did not exercise and did not diet;the diet group did not exercise and only had diet;the exercise group only exercised and did not diet;the exercise diet group exercise and diet.During the six-week experimental period,the dietary intake of each group of rats was recorded daily,and the body weight,body length,and abdominal circumference of the rats were measured weekly and the Lee's index was calculated.After the end of the experimental intervention,plasma and hypothalamus were taken.The content of NPY in rat plasma and hypothalamus was detected by double-antibody sandwich enzyme-linked immunosorbent assay.The immunohistochemical method was used to semiquantitatively study the NPY antigen of the hypothalamic arcuate nucleus.Results:(1)through dietary intervention for 8 weeks,combined with body weight,abdominal circumference and Lee rats' s index three index,selects 48 successful models of diet induced obese rats;(2)after six weeks of aerobic exercise combined with dietary intervention,the diet group rats and exercise diet group rats weight was decreased linearly,to sixth at the end of the week,were lower than the normal control group,body weight and diet and exercise group rats than diet group and exercise group;the weight of rats in six weeks in a slow upward trend,to the sixth weekend,lower than the obese control group and higher than the normal control group.The change trend of Lee and weight s index and abdominal circumference is largely the same,no weight changes obviously;(3)ANOVA and post hoc test showed that after six weeks of aerobic exercise combined with dietary intervention,diet and diet and exercise group that the plasma concentration of NPY group were higher than that of the obese control group,exercise group concentration is lower than the obese control group,after 3 hours of recovery after exercise group and exercise on plasma NPY concentration decreased,there was no significant difference;(4)after six weeks of aerobic exercise combined with dietary intervention,the diet group and exercise group is higher than the concentration of hypothalamic NPY diet in obese control group,exercise group concentration is lower than the obese control group,after 3 hours of recovery after exercise group increased hypothalamic NPY concentration,diet and exercise group hypothalamic NPY concentration decreased,there were no significant the difference;(5)after six weeks of aerobic exercise combined with dietary intervention,the expression of NPY positive cells in the hypothalamic arcuate nucleus diet group,exercise group and exercise diet group rats in three groups were lower than control group and obesity,and obesity compared to exercise immediate group control group,with significant difference(P<0.01),sports diet and exercise groups immediately compared with very significant difference(P<0.01),after 3 hours of recovery after exercise group and exercise diet group expression positive cells were increased,and the three hour exercise group and exercise immediate group compared with significant difference(P<0.01).Conclusion :(1)aerobic exercise combined with diet control can significantly reduce the body weight of rats,Lee s index and waist circumference,weight reducing effect is ideal;simple diet and exercise alone though and exercise diet control group compared the effect of a little difference,but compared with obese controls significantly effective weight loss,weight loss and diet alone is better than not only sports diet in rats.(2)the combination of aerobic exercise and dietary control experimental intervention can make the expression of pure diet rats and diet and exercise immediate rat plasma and hypothalamus NPY rise,and exercise diet group after three hours of recovery,decreased expression of plasma and hypothalamic NPY,and recovered to the normal level;(3)pure exercise can decrease the expression of plasma NPY,hypothalamic NPY and hypothalamic arcuate nucleus,and after 3 hours of recovery,the expression of hypothalamic NPY and hypothalamic arcuate nucleus increased,reaching the normal level,and the plasma NPY level decreased.(4)there is no correlation between plasma NPY levels and hypothalamic NPY concentration in plasma and hypothalamus may be due to each have a complete control system,which plays a physiological role in particular,complete the corresponding physiological function,and may be due to plasma NPY in NPY through the blood-brain barrier or other hypothalamic nuclei from the specific mechanism needs further study;(5)based on the above results,NPY in exercise combined with dietary control diet research is relatively rare,but the close relationship between NPY and exercise,obesity,diet,the NPY in the research and application of exercise combined with dietary intervention in show a very optimistic outlook,is expected to become the evaluation index criteria of obesity and weight loss however,exercise training with different loads,different ways and diet control effect and the mechanism of obesity NPY still need further research.
