Purpose: The movement has gradually became an important part of people's life in our country.Its role has aroused the attention of academics.That exercise was accepted by a lots of scholars can improve the nerve function of brain and increase learning and memory ability.However,what kind of intensity exercise can improve learning and memory ability,and whether it is related to COX5A are not clear.Therefore,this study established a treadmill model to explore the impact of different exercise intensity on learning and memory and the expression of COX5A in temporal lobe.Methods: Forty healthy male Sprague-Dawley rats(SD),aged 8 weeks,were randomly segmented into 4 groups(each group 10): control group(without training);low intensity group(18m/min);medium intensity group(24m/min);high intensity group(30m/min).The specific design scheme is as follows: training for 6 weeks,5days a week,30 minutes a day,gradient 0.we carry out the test of Morris Water maze after 6 weeks to detect learning and memory in rats.After Morris water maze test,they were banned to eat foods and drink for 24 hours,and they were injected intraperitoneally with 3.6% chloral hydrate(1ml/100g)by syringe and the temporal cortex was taken.The expression of COX5A in the temporal lobe was detected by WB and IF.Results: After 3 days of directional navigation,the average speed of the treadmill training group was lower than the control group,in which compared with control group,the low intensity group and moderate intensity group showed extremely significant difference on the first day(P<0.01),On the second day,the high-intensity group showed a significant difference compared with control group(P<0.05),on the third day,low-intensity group and the middle-intensity group showed significant difference compared with control group(P<0.01).The initial angle of four groups found three exercise groups is greater than control group on the first day,in which high intensity group was significant difference compared with control group(P<0.05),the initial angle of low intensity group was greater than highintensity group,and significant difference on the second day(P<0.05),on the third day there was no significant difference.The around distance of exercise groups was lower than control group,in which moderate intensity group was significantly different compared with control group on the first day(P<0.05),The around distance showed that high intensity group was significantly lower than low intensity group and moderate intensity group(P<0.05),On the third day,the around distance of exercise group was lower than control group,and no significant difference was found.In the one-day space exploration,frequency of passing tread of lowl intensity group and moderate intensity group increased significantly compared with control group and overall difference was not significant,but both low intensity group and the moderate intensity group had significant differences compared with high intensity group(P<0.05).Compared with the control group,the center time of treadmill training groups increased,but there was no significant difference(P>0.05).WB showed that the expression of COX5A was increased in the treadmill training group,in which low intensity group was a significant difference compared with control group(P<0.05),and the moderate intensity group was significantly different compared with control group(P<0.01).Compared with each exercise groups,the expression of COX5A in low intensity group is higher than in high intensity group(P<0.05),and the expression of COX5A in moderate intensity group is higher than in high intensity group(P<0.01).IF found that the expression of COX5A and goodness fit of neurons is higher,it is rarely expressed in glial cells.Conclusion:(1)Test index of moderate intensity treadmill training were more significant,which suggested that moderate intensity treadmill training can most likely improve the learning and memory abilities of SD rats.(2)Treadmill training can upregulate the protein expression of COX5A,especially moderate intensity treadmill training,and goodness fit is higher in temporal lobe cortical neurons.we speculated that the improvement of learning and memory abilities after 6 week moderate intensity treadmill training may more likely be related to the regulation of COX5A.
