Food safety is an important field related to people's livelihood and national security,and has been the focus of concern by the public and government departments.Food safety is an area that needs comprehensive consideration in quantity safety,quality safety,food protection,nutrition safety,and health safety,risk assessment and trade.Under the general trend of the overall good food safety situation in China,there are still many risks and hidden dangers in the food consumed by the public,which pose potential risks to the health of the public and the sustainable development of the society.From a regulatory point of view,technical supports are indispensable tools and should be sustainable development.Developing high selectivity detection methods which with high risk problem discovery rates to achieve qualitative and quantitative analysis of risk substance in food are necessary.In the face of food varieties and risk material category widespread,the database relying on the strength of science and technology need to be set up which can more relevant regulatory risk material screening of prospective demands.At the same time,in order to let the grassroots regulatory agencies play a more important role in regulatory process,the rapid food testing equipment and methods have been a typical useful tools for risk discovery.Here studies the rapid screening and detection methods of risk substances in food from three directions,and the research contents mainly include the following parts:The first part introduces the general situation of risk substances in food,summarized the common pretreatment technologies in food safety analysis,systematically introduced the rapid screening methods used in food safety analysis,and also the content and significance in current study.In the second part of this study,we developed a qualitative and quantitative method for the veterinary drug residues of imidazoles,?-receptor agonists and macrolides in milk powder and yoghurt by means of quadrupole mass spectrometry with low cost,simple operation and on-site detection.By optimizing the separation conditions of liquid chromatography,investigating the composition of extracting solvents and purification methods in pretreatment,the optimum analytical method was established.The method had good accuracy,repeatability and precision,and could be applied to on-site rapid detection because of its,small and convenient features of the instrument.At the same time,based on the problems found in the research,the direction of further analysis was put forward.In the third part of this study,based on Raman spectroscopy,a Raman spectroscopic database of more than 170 illegal compounds covering functional and non-functional components in health food was established,which contains solid raman spectra of various compounds and pH-optimized liquid surface-enhanced raman spectra.It could realize non-directional screening of prohibited substances in health food.The whole screening process could be completed in 15 minutes with short time and less reagent usage and had the advantage of rapid on-site screening.In the fourth part of this study,the first-order mass spectrometry database of more than 170 banned compounds was established by means of time-of-flight high resolution mass spectrometry,and the second-order mass spectrometry database was established by optimizing collision energy to collect abundant information of fragment ions.The databases could be used to validate the suspected positive samples screened by raman spectroscopy,or to screen the unknown samples for risk substances directly.The method was fast and accurate and compounds in the database could be updated in time with the need of supervision.The screening of samples was not limited to a specific unknown sample.It had wide applicability and was an important means to achieve effective supervision.For the purpose of rapid screening,the study used a portable mass spectrometer that was easy to implement on-site detection,and a raman spectrometer with simple pretreatment requirements and low reagent usage,and a high-resolution time-of-flight mass spectrometer to establish a method for the detection of 34 veterinary drug residues,and a raman spectroscopy database and a high-resolution mass spectrometry database of more than 170 illegal compounds in health foods,which provided a useful reference for better regulation of food safety.
