Schisandra Michx.is a perennial deciduous or evergreen woody vines attributed to the Schisandraceae.It is mainly distributed in the part of area of Northeast China,Central South and Southwest.The fruits of plants from Schisandra Michx.are used as madicine,and shape of these fruits is very similar and the plants grow staggered.The traditional identification of the plants of Schisandra Michx.are mainly relied on traditional classification,and its limitations makes identification difficult.Therefore,a simple,rapid,reliable method is needed to solve the identification and pharmacological problems.The study on the group difference of soluble protein from leaves,fruits and stems of three plants of Schisandra Michx.,S.sphenanthera,S.chinensis,S.propinqua var.sinensis by SDS-PAGE electrophoresis to provide support for identify the plants of Schisandra Michx.The main results as follows:1.By comparing the five methods for extracting soluble proteins,we found that extraction buffer 1 is most suitable for extraction in fresh leaves of S.sphenanthera,while extraction buffer 2 was most suitable for extracting soluble proteins in fruits of S.sphenanthera,S.Chinensis and S.propinqua var.sinensis.All the five methods were not appropriate for extracting soluble proteins in stems.2.By comparing the content of soluble proteins in fresh leaves of S.sphenanthera collected in different period as well as SDS-PAGE electrophoresis band,we found that leaves collected through May to July contained high content of soluble proteins,while leaves collected through July to September contained decreasing content of soluble proteins.Protein electrophoresis bands showed that seven clear bands of soluble proteins appeared in leaves of S.sphenanthera collected from May to September.No specific bands were observed although their intensity differed in bands of each period.3.To compare with three species of Schisandra Michx.plants(S.sphenanthera,S.chinensis,S.propinqua var.sinensis.)and "distinct species"(A special phenotype of S.sphenanthera)bands of soluble protein for fruit,we separated from total of 23 bands,the 7 for specific bands.We can distinguish three species of Schisandra Michx.to specific bands and to distinguish and identify them.4.The content of soluble proteins in 10 districts for 5 regions of Qinling Mountains in different years of S.sphenanthera fruits,The results show that the same year(2011),Zhashui and Ningshan with higher protein content than others of S.Sphenanthera,different year the highest Jinsixia of soluble protein in 2013,and to 2.6020 mg/g,In 2009,Fengxian collected fruit protein content was the lowest,Thereare significant differences between different collection time in the same areas of soluble protein content of S.sphenanthera fruit.Indicate the existence of regional differences in Qinling Mountains of S.Sphenanthera fruit soluble protein content,and the storage time longer,the content of protein little.5.The electrophoretic bands of S.Sphenanthera fruit collected at 10 regions in Qinling Mountains showed that there existed 7 stable common bands,and no specific bands were found.We also found that the two bands between 29.0-66.4 KDa were different in brightness,two bands of fruit collected in same year from Zhashui and Yangxian were dark.The bands of fruit collected in different years were also different.The band of fruit collected in 2013 from Jinsixia was darkest,and the band of fruit collected in 2009 from Fengxian was lightest.These data showed that the S.Sphenanthera fruit soluble protein components content are different among collections from different areas in Qinling Mountains,and when the storage time of fruit prolong,the bands of fruit change light.
