Research background:Panax notoginseng is one of the most popular traditional herbal medicines.And for its widely pharmacological functions and splendid pesticide effectiveness,Panax notoginseng was called "precious" by Li Shizhen,a master of traditional Chinese me-dicine.Mixing with others,Panax notoginseng could turn out to be plenty formulas with totally different efficients.Fufang Xueshuantong capsule is a kind of traditional Chinese medicine which was developed by the Department of Ophthalmology center of Zhongshan and Guangdong Zhongsheng Pharmaceutical Co.,Ltd.jointly.A large n-umber of long-term medical practice and experimental studies had confirmed that it w-as effective in the treatment of diabetic retinopathy,with extensive clinical application market.Recently,our group had made a series progress on pharmacodynamic constitu-ents.On the foundation,this article was about to starting with its crucial part:Panax notoginseng.We focusd on the influence in pharmacokinetics through different compat-ibility in two levels:medicines and active ingredients.This research was trying to pr-ovide some basis about the pattern of compatibility between the four medicines in Fu-fang Xueshuantong capsule.Aim:On the basis of the proportion in Fufang Xueshuantong,we were trying to figure out that,first,how the other three operate to Panax notoginseng's pharmacokinetics effects in formula between each medicine.In addition,through the reasonable compon-ent selection in formal research resently,we penetrated the influence of compatibility into ingredients' level between each active ingredient.Above all,promoted the comp-atibility effect of Fufang Xueshuantong,and provided the foundation for clarifies the chemical material basis.Contents and methods:1.Select pharmacodynamics makers in Panax notoginseng.Establish a LC-MS method to simultaneously determine the makers,and develop experimental program of pharmacody-namic.2.According to the production technology,collect each Panax notoginseng part and gather them through the ratio in compounds.Prepare separated Panax notoginseng part,which is equal to the Panax in compound,so that could apply a pharmacokinetic study of Panax notoginseng,in separation and in compounds.Finily,compare the differences between them.3.With the formula A and formula B,which were established in earlier reaserch,apply a pharmacokinetic study of Panax notoginseng's markers,in formulas and in compounds.Finily,compare the differences between them.Results:1.Notoginsenoside R1,ginsenoside Rg1 and Rb1 were selected to be the markers in pharmacokinetic study.Then a specific and sensitive LC-MS method was developed for sim-ultaneously determine.2.The normal dose of capsules for human was translated to rats by oral gavage at a dose of 1.05g·kg-1.Containing equal parts of gavage doses of Panax notoginseng,which was(0.252g extract+0.315 g powder)·kg-1.Rats were orally administered pharmacokinetic ex-periments with Panax notoginseng and capsules.Panax notoginseng,when administered alone,concenctration peaks dissipated rapidly,and the apparent volume of distribution was wider.After administration of the capsule,individual index components of Panax notoginseng,acted an increase in total body distribution,retention time increases.By compatibility,the ac-tive ingredients targets more specific in a way;besides the active ingredient and the target had a certain degree of affinity improvement.Compatibility impact on the three active ingredients in Panax notoginseng ranged ginsenoside Rb1>ginsenoside Rg1>notoginsenoside R1 in a row.3.Combine PNS with tanshinone I,cryptotanshinone,atragaloside and harpagoside side by Component prescription A,B,together with capsules four times the normal amount of 4.20g capsules·kg-1.Conducted pharmacokinetic study with rat oral administration of for-mula A and B.Notoginsenoside R1 and ginsenosides Rg1 apparent pharmacokinetic parame-ters in formulas were significant differences in the capsules.Ginsenoside Rb1,presented from A to B with prescription capsules showed a trend that close to the capsule by prescription compatibility.Conclusion:Through the compound compatibility of the four herbs in Fufang Xueshuantong,the ac-tive ingredients of Panax notoginseng were significant impact,such as released slowly and solubilization.Through the compound compatibility of formulas,the markers show signifi-cant differences with the capsules.And show a trend that close to the capsule by prescription compatibility.
