Objective:To investigate whether Zhenwu Decoction regulates cardiomy-ocyte apoptosis through affecting the expression of miRNA-1 and miRNA-133 in heart and kidney yang deficiency CHF rats.Methods:Fifty SD rats were randomly divided into 3 groups(10 in the sham group,20 in the model group,and 20 in the Zhenwu Decoction group),and males and females in each group accounted for half.Rats in the model group were resected with bilateral thyroid gland.10 days after surgery,rats were injected intraperitoneally with 2mg/kg body weight of doxorubicin(2mg/ml with water for injection,ie,0.1 ml/100 g body weight),twice a week for 3 weeks.Rats in the sham group cut the skin,strip muscles,expose the thyroid,and then suture muscles and skin.10 days after surgery,rats were injected intraperitoneally with normal saline 0.1ml/100 g body weight twice a week for 3 weeks.After the model was completed,the Zhenwu Decoction group was given to the stomach by Zhenwu Decoction(4.26g/(kg·d)of equivalent piece)once a day for 8 weeks.At the same time,the sham group and model group were given equal doses of saline to the stomach once a day for 8 weeks.At the end of treatment,the heart function and hemodynamics of rats were measured.HE staining was used to observe the myocardial cells,the left ventricle miRNA-1 and miRNA-133 expression were detected by Real·time PCR,the expressions of Caspase-3 and Caspase-9 were detected by immunohistochemistry and TUNEL assay Tissue apoptotic rate.Results:(1)Zhenwu Decoction can improve the heart function of CHF rats with heart and kidney yang deficiency and increase the LVEF and LVFS values.(2)Zhenwu Decoction can improve the hemodynamics of heart-kidney yang deficiency CHF rats,down-regulate LVEDP,increase LVAP and ±dp/dtmax.(3)Zhenwu Decoction can improve the body structure of myocardial cells in CHF rats with heart and kidney yang deficiency.(4)Zhenwu Decoction can decrease the Apoptosis Rate of Myocardium in CHF Rats with Heart-Kidney Deficiency.(5)Zhenwu Decoction has no significant effect on the expression of miRNA-1,miRNA-133,caspase-3,caspase-9 in cardiomyocytes of CHF rats with heart and kidney yang deficiency.Conclusion:Zhenwu Decoction can inhibit cardiomyocyte apoptosis and improve heart function and hemodynamics in heart and kidney yang deficiency CHF rats.However,it did not affect cardiomyocyte apoptosis by affecting the expression of miRNA-1 and miRNA-133 in cardiomyocytes.The related mechanism of action remains to be further studied.
