The Effect Of Shennao Fuyuan Decoction On EPO In Rats With Cerebral Ischemia And Reperfusion Was Explored Based On The Theory Of "Shen And Brain Simultaneous Treatment"

Objective: Observe the intervention effect of Shennao Fuyuan Decoction on the expression of EPO,EPOR,HIF-? in the brain tissue of rats with cerebral ische mia-reperfusion,and speculate the basic biological relationship of kidney brain.It provides a theoretical basis for the clinical application of the "kidney-brain treatment" method to reduce neurological deficits and neuronal damage and improve prognosis.Methods: Forty SD rats were randomly divided into 4 groups using the random number method: blank group,sham operation group,model group,and Shenna ofuyuan decoction group.The model was used to establish the focal cerebral ischemia model of middle cerebral artery infarction.The Shennaofuyuan decoction group was diluted with Shennaofuyuan decoction,and the other groups were given the same amount of distilled water.After 3 days of gavage,the animals were killed and the materials were taken.HE staining was used to observe the pathological changes of brain tissue.Immunohistochemical method was used to detect the expression of HIF-?,EPO and EPOR.rt-PCR was used to detect the expression of HIF-?,EPO and EPOR m RNA in the peripheral area of cerebral in farction.Results:1.Condition of rats: Shennao Fuyuan Decoction can improve the symptoms of neurological deficits in rats.2.Immunohistochemical analysis of HIF-?,EPO,EPOR protein: Compared with the model group,the Chinese medicine group HIF-?,EPO,EPOR showed a high expression trend(P<0.05).3.rt-PCR analysis of HIF-?,EPO and EPOR m RNA: Compared with the model group,HIF-?,EPO and EPOR m RNA were increased in the Shennaofuyuan Decoction group(P<0.05).Conclusion: When cerebral ischemia is perfused in rats,intervention of Shennaof uyuan decoction can promote high expression of HIF-?,which in turn stimulates EPO production,EPO feedback promotes EPOR expression,and EPO plays a role in protecting neurons by combining with EPOR.There is indeed a correlation between EPO and kidney essence in terms of theoretical connection and physiological relationship.Through Yishen filling the marrow,Shenxue Fuyuan Decoction of Huoxue Tongluo can increase the expression of endogenous EPO in rat brain tissue.One of the material foundations.
