Flos Datura is the dry flower of Datura metel L which belongs to the family of Datura Linn.,known as ’Naoyanghua’,’Fengqiehua’,’Labahua’ and ’Mantuoluohua’,and it is widely distributed in Guangdong,Jiangsu,Zhejiang and Fujian provinces of China.As a traditional Chinese medicine,D.metel L.has a long history of medicinal use.D.metel L.has been used clinically for the treatment of asthma,cough,pain,rheumatism,convulsions,anesthesia,etc.The chemical constituents of D.metel L.mainly include withanolides,alkaloids,flavonoids,terpenoids and lignans.Modern pharmacological studies have shown that D.metel L.has an effect on the central nervous system,and a wide range of pharmacological applications,such as antitumor activity,antiinflammatory activity,analgesia,antimicrobial activity,antioxidation activity,and hypoglycemic activity.Magnoliae officinalis Cortex is a dry skin,root bark and branch bark of Magnolia officinalis Rehd.et Wils.and Magnolia officinalis Rehd.et Wils.Var.biloba Rehd.et Wils.,and has been extensively used as a traditional Chinese medicine for clinical treatment.Chemical study was focused on lignans,phenylethanoid glycosides,phenols,alkaloids,etc.Pharmacological studies have shown that M.officinalis is mainly for the protection of nerves,cardiovascular and cerebrovascular,and it has good activities in antioxidation,antibacterial,antiinflammatory,anti-ulcer,antitumor,etc.In order to clarify the pharmacodynamic basis of D.metel L.and M.officinalis,this paper reported systematically the isolation and identification a series of compounds,evaluated withanolides isolated for antitumor activities,and screened lignans for antioxidant activities to provided scientific basis for the further development and utilization.In the first part of this paper,we have researched systematically the chemical constituents of 95%ethanol and 70%ethanol extracts from D.metel L.and a total of 22 compounds were isolated by various chromatographic separation methods(silica.,gel,ODS,PHPLC,ect).These chemical structures of all the isolates were established by extensive spectroscopic analysis(UV,IR,HRESIMS,NMR,ECD,X-ray crystallography),including 10 withanolides,4 amides,5 terpenoids,and 3 other compounds.The structures of these 22 compounds were characterized as follows:daturanolide B(1*),daturanolide A(2*),daturanolide C(3*),withametelin C(4),withatatulin(5),withametelin G(6),lycium substance B(7),12-deoxywithastramonolide(8),withametelin E(9),withafastuosin A(10),aristopyridinone A(11),aurantiamide(12),aurantiamide acetate(13),scutebarbatine A(14),(+)-dehydrovomifoliol(15),blumenol A(16),(6R,7E,9R)-9-hydroxy-megastigma-4,7-dien,3one(17),oleracone B(18),isololiolide(19),P-hydroxybenzene propanoic acid(20),(-)-syringaresinol(21),carisphthalate(22).Among them 3 new compounds(1-3)were firstly reported,daturanolide A(2)represented a rare example of withanolides with a 2-en-1,6-dione A/B ring system,and 12 compounds(11-22)were firstly found in Datura Linn.By u sing MTT assay,withanolides(1-9)were evaluated for their cytotoxicity on HCT116,U87-MG,NCI-H460,BGC823,and HepG2 cell lines,and 3,6,7,8 exhibited good inhibitive activities.From the above results,it was deduced that the formation of a C-21-O-C-24 ether bridge,an exocyclic double bond at C-25-C-27,6,7-epoxides,and 2,4-dien-l-one system could enhance antitumor activities of withanolides,as well as the 12-OH structure may impair the antitumor activities.In the second part of this paper,we studied the chemical constituents and antioxidation activities of M.officinalis.13 compounds were isolated and confirmed from 95%ethanol extract from M.officinalis as follows:magnolol(1),honokiol(2),randainal(3),magnaldehyde B(4),magnaldehyde E(5),streblusol E(6),magnatriol B(7),randainol(8),magnolignan C(9),2-[2-(hydroxymethyl)-1-benzofuran-5-yl]-4-(prop-2-en-1-yl)phenol(10),syringaldehyde(11),cryptomeridiol(12),2,6,2’,6’-tetramethoxy-4,4’-bis(2,3-epoxy-l-hydroxypropyl)biphenyl(13).Among them 10 and 13were first discovered in M.officinalis.The antioxidant activities of lignan compounds 1-8 were investigated by DPPH method.The results showed that these lignan compounds have certain antioxidant effects.The structure-activity relationship study indicates that the ortho or para position of phenol is introduced into the allyl group,which can enhance the scavenging activity of free radicals;the number of allyl groups affects the rate of free radical scavenging inhibition.
