Studies On Chemical Constituents Of Active Parts For Paoriasis In Datura Metel L.

Flos Datura is dried flower of an annual herb Datura metel L. which belongs to the family of Solananeae. And it is a traditional herb.It has been recorded in 2005 edition of 《Chinese Pharmacopoeia》 .It has been used as folk herbal drug in China and it has high bioactivities and wide utilization.It can relieve cough and asthma;alleviate pain and convulsion, etc. The Drug has been used for the treatment of psoriasis in these years.It showed an excellent curative effect with many advantages .But it had obviously negative effect Our team have studied the chemical constituents against psoriasis and pharmacological effects in these years. We found and confirmed that the chemical constituents extracted with 50% ethanol from Flos Datura was the effective part for the treatment of psoriasis . we also found the effective part has include Flavonoids and Withanolide compounds, this article deals with the chemical constituents in effective part of treatment.In our studies on chemical constituents in effective parts of treating psoriasis, 8 compounds were isolated with silica, gel , reversed-phase (ODS) , Sephadex LH-20 column chromatography and HPLC. The structures of these 5 compounds were fully determined according to the physicochemical and spectrum evidence (1D and 2D-NMR). 5 novel compounds were firstly reported . Among them there are 2 sesquiterpenoids compounds and 3 withanolide compounds, these five compounds were named as DaturametelosideN[(3S,5R,6R)3,5,6,7-tetrahydroxy-Megastigmane-7-en-9-oxo-3-O-β-D-glucopyranosid],Daturameteline N[(3S,5R,6R)3,5,6,7-tetrahy-droxy-megastigmane-7-en-9-oxo], Daturameteline K[5α,6β,12β,21,24,29-hexahydroxy- (20R,22R-l-oxo- witha-2-enolide), Daturametelline L[5α, 60,12β,21,24, 25,29 -hetahydroxy - (20R,22R) -1-oxo- witha-2-enolide] and Daturameteline M [5a,6β,12β,27-tetrahydroxy-21,24-epoxy-(20S,22R, 24R,25R)-1 -oxo-witha-2-en-olide ].This thesis further reserched the chemical constituents of effective part inFlos Datura and the effective substances for the treatment of psoriasis. All these will provide important scientific foundation for further reserch and the development of new drugs and will promote the study.
