Studies On Chemical Constituents Of Datura Metel.L In Effective Parts Of Treating Psoriasis

Flos Datura is dried flower of an annual herb Datura metel L. which belongs to the family of Solananeae. It was recorded originally in 《Compendium of Materia Medica 》 and has been recorded in 2005 edition of 《Chinese Pharmacopoeia》 . It has been used as folk herbal drug in China and it has high bioactivities and wide utilization.It can relieve cough and asthma;alleviate pain and convulsion, etc. The injection of Datura has been used for the treatment of psoriasis these years.It showed an excellent curative effect with many advantages such as high cure rate , low relapse rate and so on.Recently there have been many reports about its chemical constituents, pharmacological effects , clinical studies .However the chemical constituents against psoriasis have not been stuided and found yet. In order to elucidate its action mechnism against psoriasis,this article deals with the chemical constituents in effective part of treatment.In our studies on chemical constituents in effective parts of treating psoriasis, 11 compounds were isolated with silica, gel, reversed-phase (ODS), Sephadex LH-20 column chromatography and HPLC. The structures of these 11 compounds were fully determined according to the physicochemical and spectrum evidence (1D and 2D-NMR). 2 novel withanolide compounds were firstly reported and 3 compounds were firstly found in Datara metel L .Among of them there are 5 withanolide compounds, 3 lignans compounds, and 3 others.The structures of 5 withanolide compounds were identified as Daturameteloside G(5α,6β,27-trihydroxy-(20R,22R)-1 -oxo-witha-2,24-dieno lide-27-O-β-D-glucopyranoside),Daturameteloside H(5α,6β,12β,27-tetrah ydr oxy- (20R,22R) -1-oxo- witha-2,24-dienolide- 27-O-β-D-glucopyranoside) , WithafastuosinF,WithafastuosinE,WithametelinC,among of them Daturamet -elostde G and Daturameteloside H are novel compounds; the structures of 3 lignans compounds were identified as (+)-isolariciresinol, (+)-pmoresinol-β-D - diglucoside and (+) -pinoresinol-β-D-diglucoside.The result of this research provides important scientific foundation and material base for further research of bioactivities,pharmacological action...
