of HCC.ObjectiveThis experiment by HCC precancerous lesion of rat model study,by intraperitoneal injection method of diethyl-nitrosamine lesion model in rats,By giving the object in different moxibustion on gan shu acupoint therapy,Observation of direct moxibustion and gan shu acupoint on rats hepatic effects of TSGF,Livin,Explore the value of direct moxbustion-Gansh u point in role in HCC development in rats and the action mechanism.MethodsGrouping and modeling methods:A total of 70male Wistar rats were randomly divided into control group(n=10),model group(n=15),direct moxibustion-single group(n=15),direct moxibustion-double group(n=15)and ginger moxibustion-single group(n=15).HCC model group was established by intraperitoneal injection of diethylnitrosamine(DEN,50mg/kg),once every 3dys for 10 weeks.Direct moxibustion-single group and direct moxibustion-double group were respectively applied to bilatreral "Ganshu"(BL 18)with 3 seed-sized-moxa cones or 6seed-sized-moxa cones,ginger moxibustion-single group was treated with 3 seed-sized moxa cones,once every other day,the same as the model group's treatment.Rats in the normal group received no treatment.After finishing the establishment of the model,the rats were made anesthesia with 10%of chloral hydrate,the liver tissues and blood were taken for detection.After cutting out the whole liver,weighted the liver,then calculated the liver index;The liver with hepatic lobule were removed for HE staining so that the pathological morphological changes of liver tissues were observed;Using the method of ELISA detected the expression of serum TSGF.The epression of the inhibitor of apoptosis-livin in the liver tissue was detected with immunohistochemical method.Results1.The pathological morphology change of the liver tissue:HE staining showed that Uneven cell size of model group,Cancerous cells with hyperchromatic,Shaped nodules,mitoses,absent a central vein,collagen,hepatic sinusoids are too narrow,there are megakaryocytic cells,Heparin arranged disappear.In the moxibustion group,Cell staining is equal,lower cellular pleomorphism,nodule formation less liver tissue with group have a certain degree of improvement.In the normal group,liver cells have normal,liver arranges,hepatic trabecular small,liver is not abnormal.2.The expression of serum TSGF:After establishing a HCC rat model,compared with the normal group,the content of serum TSGF increased oviously(P<0.05),Compared with the model group,in the direct moxibustion-single group and direct moxibustion-Joule group,the content of serum TSGF ruduced(P>0.05).Compared with the model group,in ginger moxibustion-single,the content of serum TSGF reduced obviously(P<0.05).3.The expression of Livin in the liver tissue:After establishing a HCC rat model,Compared with the normal group,in the model group,direct moxibstion-single group and direct moxibustion-doule group and ginger moxibustion-single group,The expression of Livin increased obviously(P<0.05).Compared with the model group,in the direct moxibustion-sin gle group,The expression of Livin decreased(P>0.05),in the direct moxibustion-double group and ginger moxibustion-single group,the expression of Livin decreased obviously(P<0.05).Conclusion1.In the study,we used the DEN method to induce the establishment of HCC rat model,and the method was reliable.2.Direct moxibustion and ginger moxibustion on Ganshu point can improve liver pathol ogy of HCC Rat model and reduce the level of cancer.But no difference in improvement betw een direct moxibustion and ginger moxibustion.3.Direct moxibustion on Ganshu point can reduce effectively the expression of TSGF an d Livin..4.Direct moxibustion Ganshu point of HCC rat model has a certain degree of preventive effect,not completely blocked,but has the ability to slow down the cancer process.Direct moxibustion Ganshu point can redcue the expression of TSGF and Livin,which has Inhibition of cancer cell formation.
