Study On The Extraction Process And Antioxidant Activity Of 5-hydroxymethyl Furfural, A Medicinal Component Of Shengmai Powder

Objects:To explore the effect of different pH solvents on the dissolution efficiency of active ingredient 5-hydroxymethylfurfural(5-HMF)in Shengmaisan,and optimize the extraction process of Shengmaisan;To explore the antioxidant activity of Shengmaisan and 5-HMF,and prove that the optimized extract of Shengmaisan has The pharmacological action against oxidation of the body.Methods:1.Extraction process of Shengmaisan(1)Using the content of 5-HMF as the index,the single extraction method was used to determine the optimal extraction solvent,the optimum solvent amount,the optimal soaking time,the optimal extraction time and the optimal extraction times.(2)Based on the single factor experiment,the optimal extraction process of Shengmaisan was optimized by four-factor and three-level L9(34)orthogonal test method.2.Shengmaisan antioxidant activityThe antioxidant activity of the optimized extract of Shengmaisan was evaluated by measuring the clearance rate of 5-HMF to ABTS~+radical and DPPH radical by ABTS method and DPPH method.Results:1.Schisandra does provide the role of acidic environment in the prescription.The acidic environment is the key to the dissolution of active ingredient 5-HMF in Shengmaisan.The optimal extraction solvent for Shengmaisan is 5%acetic acid.2.Through the single factor experiment and the four-factor three-level orthogonal test results,the optimal extraction process of Shengmaisan is 10 times of solvent addition,soaking for 60min,extracting 1.5h,extracting twice;Using this optimal extraction process to extract Shengmaisan,the extraction rate of the active ingredient 5-HMF in Shengmaisan was 6.85%.3.The experiment of antioxidant capacity test showed that Shengmaisan acetic acid extract and 5-HMF have some antioxidant capacity,which proves that Shengmaisan acetic acid extract has antioxidation pharmacological effects,these can provide some theoretical basis for the clinical treatment of diseases of body oxidation.Conclusions:The acidic solvent is beneficial to the dissolution of the active ingredient 5-HMF of Shengmaisan;the optimal extraction process of Shengmaisan is to use 5%acetic acid as solvent,soaking solvent 10 times,soaking for 60min,extracting 1.5h,extracting twice;5-HMF has some antioxidant capacity,Shengmaisan does have antioxidant pharmacological effects,and these can provide some theoretical basis for the clinical treatment of diseases of body oxidation.
