Purpose: In the established mouse model of diabetic skin ulcer,the application of Yixiao cream was used to observe the effect of Yixiao cream on the repair and healing of wounds.The mechanism of promoting the repair and healing of diabetic skin ulcers was preliminarily explored for further research and clinical application.Material and method:28 SPF Kunming male mice were randomly divided into two groups according to the random number table: 21 diabetic skin ulcers and 7 normal groups.After successful modeling,the diabetic skin ulcers were randomly selected.The method was randomly divided into three groups: Yixiao cream group;rehabilitati on new fluid group;normal saline group.The three groups were treated with Yixiao cream;rehabilitation new fluid;normal saline was changed at the wound daily,and the dressing was changed continuously for 14 days.The general state of the experimental mice was observed daily,and the size of the wound,the amount of exudation and the granulation of the newborn were closely observed.On the 14 d,the wound area of each mouse was measured by the thin film method,record wound area and calculate healing rate.The wounds were taken at the wound surface,HE staining and immunohistoch emistry were used to observe neonatal capillaries;fibroblasts;inflammatory cell number changes,proliferating cell nuclear antigen(PCNA)and insulin-like growth factor-1 receptor(IGF-1R)expressionand statistical analysis was performed using statistical softw are SPSS 17.0.Results: 1.After adaptive feeding,there was no significant difference in fasting blood glucose and body weight between the groups.The fastingblood glucose of the model mice before the dressing change was significantly higher than that of the normal group(P<0.05).During the dressing change,the Yixiaocream group,the rehabilitation new fluid group,the normal saline group showed weight loss,lack of spirit,and the skin was owed gloss,the reaction is slightly retarded,the amount of food and water is obviously increased,the amount of urine is large,the color is yellow and thick,and the smell is odorous.The fastingblood glucose and body weight of the mice in each group were measured 14 days after dressing change.There was significant difference between the normal group and the model group(P<0.05),but there was no significant difference between the three groups(P>0.05).2.The wound of the Yixiao cream group was moist,the base color was red,and the area was shortened quickly;the rehabilitation new liquid group had exudation when the wound surface was dark,the base color was dark red,and the area was shortened faster;the saline group had more exudation and the base color dark,the area shrinks slowly.On the 14 th day of dressing change,the area of wound reduction was measured and the healing rate was calculated: the area of wound reduction and healing rate of Yixiao cream group were higher than that of rehabilitation new fluid group(P<0.05);the area of wound reduction of Yixiao cream group and rehabilitation new fluid group and The healing rate was better than that of the saline group(P<0.05).HE staining of the basal tissue of the wound showed that a large number of new capillary capill aries inflammatory cells were observed in the section of the first-effect cream group.Some new capillary capillaries were observed in the section of the rehabilitation new fluid group,and a small amount of inflammatory cells still existed.In the saline group,there were few new capillaries and a large number of inflammatory cells infiltrated.No new capillary and inflammatory cells were seen in the normal group.Immunohisto chemical examination showed that the expression of IGF-IR and PCNA was statistically analyzed: the expression of IGF-1R in the saline group was lower than that in the normal group(P<0.05);the expression of PCNA in the saline group was higher than that in the normal group((P<0.05));the expression of both in the Yixiao cream group was higher than that in the rehabilitation new fluid group(P<0.05).Conclusion: 1.Yixiao cream can promote the healing of experimental diabetic skin ulcer wounds.2.One of the mechanisms of Yixiao cream to promote the healing of experimental diabetic skin ulcers is to up-regulate the expression levels of PCNA and IGF-IR in tissues.
