Paranoid personality disorder(PPD)is one of the common personality disorders.It is caused by a variety of genetic and environmental factors.Childhood abuse is generally considered to be an important factor.Nitric Oxide Synthase 1 Adaptor Proteins(NOS1AP)are potent candidate genes for schizophrenia,and PPD has similar physiological and pathological basis with schizophrenia.Therefore,this study explored the regulatory effect of NOS1AP gene polymorphism between childhood abuse and PPD in male violent crimes in Chinese Han population.This study used convenience sampling,and 495 male inmates of violent criminals in a prison in Nanjing were divided into PPD group and non-PPD group acessed by Personality Diagnostic Questionnaire-4(PDQ-4).Exclude participates who have serious physical illnesses and other mental disorders,and who are not Chinese Han people.All the participates were evaluated by the self-compiled general demographic data questionnaire,family information questionnaire,substance use questionnaire,PDQ-4,Generalized Anxiety Disorder-7(GAD-7),Patient Health Questionnaire-9(PHQ-9),Childhood Trauma Questionaire-28 item Short Form(CTQ-SF),Barratt Impulsiveness Scale(BIS-11)and Self-rating Idea Of Suicide Scale(SIOSS).All subjects took 5 ml of whole blood,extracted DNA,and do genetic polymorphism detection.Statistical analysis was performed using SPSS 25.0,and the scales and genetic data of the two groups were compared.Main results are as follows:(1)Significant difference were found between the PPD group and the non-PPD group in paternal rearing style(?2=9.199,P=0.027)and parental relationship(?2=8.077,P=0.044).The scores of anxiety,depression,suicidal ideation,motion impulsivity,unplanned impulsiveness,impulsive total score in the PPD group were higher than those in the control group significantly(P<0.05).(2)Bivariate Pearson correlation analysis showed significant correlation between PPD scores and emotional abuse,physical abuse,sexual abuse,emotional neglect,physical neglect,and CTQ total score(P<0.05).But partial correlation analysis showed that only emotional abuse was significantly associated with PPD scores(r=0.139,P=0.004).Significant difference were found between the PPD group and the non-PPD group in emotional abuse,physical abuse,emotional neglect,physical neglect,and childhood abuse(P<0.05).Stepwise regression analysis showed that emotional abuse had a significant predictive effect on PPD(P=0.004).(3)One-way anova analysis showed that the PPD scores of the three genotypes of rs348624 and rs4145621 were significantly different(F=3.184,P=0.043;F=3.541,P=0.030).Regression analysis showed that the SNPs rs348624 and rs4145621 entered the equation.(4)The results showed that the SNP of NOS1AP,rs3751284,has a significant regulatory effect between emotional neglect and PPD(R2=0.0136,t=-2.042,P=0.042).The SNP rs4145621 modulated the total score of childhood abuse and PPD score(R2=0.0189,t=-2.468,P=0.014).The SNP rs6680461 had a significant regulatory effect on the total scores of emotional,physical,sexual and childhood abuse and PPD scores(R2=0.023,t=2.771,P=0.006;R2=0.031,t=3.17,P=0.002;R2=0.026,t=2.822,P=0.005;R2=0.016,t-2.223,P=0.027).Main conclusions are as follows:(1)Childhood abuse experiences,especially emotional abuse,has close correlation with PPD symptoms among violent crime male prisoners;(2)NOS1AP gene,SNP rs348624 and rs4145621,have close correlation with PPD symptoms among violent crime male prisoners;(3)The interaction between NOS1AP gene rs3751284,rs4145621,rs6680461 polymorphism and childhood abuse plays an important role in the pathogenesis of PPD among violent crime male prisoners.
