In this paper,we propose to develop medicinal bath agent by using chlorine dioxide as active ingredient sterilization agent,which including two effective components,group A and group B.Group A is compound sodium chlorite effervescent tablets and group B is film-forming substrate.The physicochemical indexes and stability of two groups were respectively investigated.In addition,the microorganism sterilization experiment and skin irritation experiment of cow nipple bath were also carried out.The main contents are shown as follows:The single factor and orthogonal test of group A and group B were respectively carried out to obtain the prescription of group A(compound sodium chlorite effervescent tablet)including 22% sodium chlorite,50% citric acid,8% sodium bicarbonate,5%polyethylene glycol 6000 and 15% anhydrous magnesium sulfate.The prescription for the group B(film forming matrix)is 2% sodium alginate in 100 mL,4% plasticizer,2%skin-care agent and 5% surfactant.The 1 piece of compound sodium chlorite effervescent tablets was activated in a closed container containing 50 mL of water for 10 min,and then diluted 10 times to obtain A solution with a chlorine dioxide content of about 100 mg/L.At last,solution A and group B were mixed by 1:1,stirred evenly to obtain cow nipple bath.Through the physicochemical indexes of group A,the properties,p H,disintegration time limit,weight difference,friability and hardness all meet the requirements of Chinese Veterinary Pharmacopoeia.The traits,p H,and stability of group B also meet the requirements of the Chinese Veterinary Pharmacopoeia.The film formation time is short and the film adhesion is strong.Moreover,the membrane has good air permeability,which meets the relevant quality requirements of film-forming matrix.The iodometric method was used to determine the content of active ingredients in medicinal bath agent and the methodological study was conducted.The results showed that the method has high accuracy,good precision,good repeatability.It can be used to detect the content of active ingredients of the product.The bactericidal performance of medicinal bath agent was well.When the content of chlorine dioxide was 50 mg/L and the bacteria were used for 5 min,the staphylococcus aureus and escherichia coli were completely killed.In this study,the advantages of medicinal bath agent prepared were good sterilization effect and high safety.No erythema and edema was found in the skin irritation test,indicating it has no irritation and nontoxicity.The bath medicine could be used in the clinical trials of dairy cows.The stability test showed that group A is not sensitive to high temperature and strong light,but it is very sensitive to high humidity environment.Meanwhile,accelerated and long-term experiments show that the use of aluminum foil composite bags as a packaging material can guarantee the stability of the group A against moist heat.Finally,long-term experiments exhibited that the stability of the group B was still good after 18 months.Similarly,group B also has good stability in high temperature,strong light,and high humidity,etc.There is no significant change in each index under accelerated and long-term test conditions.Furthermore,the stability of cow nipple bath was investigated,the validity period can reach 7 days under the condition of confined and protected from light.
