| Fuyang lansi radish is a local characteristic variety,but in recent years,the variety has deteriorated seriously and the quality is uneven,so it is in urgent need of improvement.Chemical mutagenesis breeding can obtain rich germplasm resources in a short period of time.Ethyl methyl sulfonate(EMS)is widely used in mutagenesis breeding of common crops,including radish,but there is no research on the characteristic variety of Xisi radish in Fuyang.In order to improve the quality of Fuyang Lansi radish and breed excellent varieties,the effects of EMS on seed germination and agronomic characters of Fuyang Lansi radish were studied.In this experiment,different concentrations of EMS(0,0.2%,0.4%,0.6%,0.8%)were used to soak the seeds of Fuyang Lansi radish.The treatment time was 4h,6h,8h,respectively,to study the mutagenesis effect of EMS on the germination potential,germination rate,seedling height and root length of Fuyang Lansi radish.With half lethal concentration as the EMS concentration selection standard,we soaked Fuyang Lansi radish in 0.6% EMS concentration solution for 4h and 6h.Through field experiment,we investigated the field agrarian characters and fleshy root quality of Fuyang Lansi radish,further clarified the mutagenic effect of EMS on Fuyang Lansi radish,and looked for excellent mutant lines.The main research work and achievements are summarized as follows:(1)The experimental results showed that with the increase of EMS concentration(0.2%~0.8%),the seed germination rate,root growth and seedling growth of Fuyang Lisi radish all showed a decreasing trend,and the inhibition effect of EMS on root was stronger than that on seedling.Under the same EMS concentration,with the increase of treatment time,the germination rate of Fuyang Lansi radish seeds showed a decreasing trend,and the germination time was delayed.(2)At present,many studies have determined that the optimal dose is 50% seed germination,that is,half lethal dose.In this experiment,the optimal concentration of EMS treatment of Fuyang Lansi Radish seeds is 0.6%,and the treatment time is 6h.(3)The Fuyang Lansi radish treated with half lethal dose was sown in the field,and the diversity and mutation frequency of the mutant traits such as leaves,stems,flower organs and fleshy root quality of M2 population were investigated and analyzed,laying a solid foundation for the establishment of the mutants induced by Fuyang Lansi radish in the future. |
