| By utilizing the characteristics of the carnivorous plant parasitic on the roots of desert plants,Cistanche can be cultivated through large-scale construction of desert plantations,and economic income can be realized while realizing ecological improvement in the desert area,achieving ecological and economic "win-win".However,due to the poor understanding of the quality,germination characteristics and the process of parasitic to desert plants,the artificial cultivation is mainly realized by simulating natural parasitic processes,which not only has the problems of large amount of seeds and low germination rate,but also high cultivation cost and unstable production.This study,the liquid inoculation technique of Cistanche was used as the target.Field sampling,laboratory analysis and field experiment were used to analyze the seed quality and germination characteristics of the different parts of Cistanche inflorescence and the field test of liquid inoculation method.Cultivation from seed collection to inoculation provides a complete technical solution.Below are key research findings:(1)The lower part of the Cistanche inflorescence(1/3 inflorescence length)had the best seed quality,the middle part(1/3 inflorescence length)followed,and the upper part(1/3 inflorescence length)was the worst.The lower part of the inflorescence has a thousand-grain weight of 0.099 g,a viability of 81.21%,a germination rate of 79.98%,and a seed particle size of more than 0.5 mm by weight,accounting for 88.14%.(2)The four indicators of germination rate,fullness,particle size and 1000-grain weight can be used as the main grading indicators for judging the quality of the seeds of Cistanche Grade Ⅰ and Ⅱ seeds are relatively high quality seeds,which can meet the requirements of seed quality in the production of Cistanche.The quality of seeds of Grade III and below is poor and cannot meet the requirements of seed quality for production.(3)A mixture of 10 mg·L-1 fluridone and 100 mg·L-1 gibberellin(GA3)can effectively promote the germination of Cistanche seeds,and the germination rate is over 70%,and the germination is neat.(4)With a nozzle diameter of 2.0 mm,the spray volume is moderate,which can effectively shorten the distance between the seed and the host root system and increase the contact area between the seed and the host root system.The best ratio of seed inoculum 2,6-dimethoxy-p-benzoquinone(2,6-DMBQ)0.02 mg·L-1 humic acid 30 g·L-1,rooting powder(ABT)50 mg· L-1,the inoculation rate can reach more than 75%.(5)The liquid inoculation method is not only easy to operate,but also has a small amount of seeds,and can effectively increase the inoculation rate of Cistanche,and achieve stable production and high yield of artificial cultivation. |
