ObjectiveWuzhimaotao(五指毛桃)and Xianhecao(仙鹤草),the lingnan traditional Chinese medicine,have the function of regulation the spleen and stomach.However,there are few researches on its mechanism of action.Therefore,this study observe Wuzhimaotao and Xianhecao(aqueous extract)on the prevention and treatment of the injury of gastrointestinal mucosa caused by indometacin and ethanol in rats.In addition,the protective mechanism of gastrointestinal mucosa was discussed from the perspective of regulation of tight junction proteins,adhesion junction proteins and transcription factors.Methods(1)To extract the aqueous extract of Wuzhimaotao and Xianhecao.Wuzhimaotao or Xianhecao was soaked in 12 times of water for 2 hours,boiled for 2 hours,filtered,and fried with 10 times of water for 2 hours,filtered.Two filtrates were combined,and then rotated-evaporation,freeze-drying,and freeze-dried powder were collected.(2)Replication of indomethacin induced gastric mucosal injury in rats.The animals were adaptively fed for 7days and fasted but water allowed for 24h,indomethacin(40mg/kg)was injected subcutaneous at 10ml/kg.After fasting and water deprivation for 7h,the animals were sacrified to observe the gastric mucosal damage.(3)Replication of ethanol induced gastric mucosal injury in rats.The animals were adaptively fed for 7days and fasted but water allowed for 24h,gavaged with anhydrous ethanol at 5ml/kg.After fasting and water deprivation for 1h,the animals were sacrified to observe the gastric mucosal damage.(4)Study on the prevention of Wuzhimaotao and Xianhecao to the gastric and intestinal mucosal injury induced by indomethacin and the gastric mucosal injury induced by ethanol in rats.(5)Pathological sections of HE staining were used to observe the gastric mucosa damage.(6)Immunohistochemistry were carried out to detect the expression of tight junctions proteins(Claudin-1、Occludin、ZO-1),adhesion junctions proteins(E-cadherin、α-catenin、β-catenin)and transcription factor SOX2 和 CDX2 in gastric mucosa.Results:(1)The freeze-dried powder of the aqueous extract of Wuzhimaotao and Xianhecao and its spectrum results:The yield of freeze-dried water extract of Wuzhimaotao and Xianhecao was 11.6%and 15.4%,respectively.The same absorption peaks of psorden(补骨脂素)and apogenin(芹菜素)in standard preparation were also detected in Wuzhimaotao decoction.The retention time of psorden and apogenin was 57.838min and 66.130min,respectively.(2)The influence of Wuzhimaotao to the gastric mucosal injury induced by indomethacin and ethanol in rats:Wuzhimaotao(14g/kg)can prevent and cure indomethacin-induced gastric mucosa injury in rats(their gross score compared with model group,P<0.05).However,could not prevent the mucosal damage of stomach cause by ethanol.(3)The influence of Xianhecao to the gastric mucosal injury induced by indomethacin and ethanol in rats:Xianhecao(16g/kg and 8g/kg)can prevent and cure ethanol-induced gastric mucosa injury in rats.However,could not prevent the mucosal damage of stomach by indomethacin.(4)Changes of related proteins in indomethacin and ethanol model rats.In indomethacin and ethanol model rats,the expression of gastric mucosa tight junction proteins(Claudin-1、Occludin、ZO-1),adhesion junction proteins(E-cadherin、α-catenin)and transcription factor SOX2 decreased,but there were no significant change in the expression of adhesion junction proteinβ-catenin and transcription factor CDX2.(5)Effects of Wuzhimaotao on indomethacin model rats tight junction proteins,adhesion junction proteins and transcription factors.In the indomethacin model rats,Wuzhimaotao had an improvement effect on the decrease of gastric mucosal tight junction proteins(Claudin-1、Occludin、ZO-1),adhesion junction proteins(E-cadherin、α-catenin)and transcription factor SOX2(P<0.05 compared with the model group),but had no significant effect on the expression of β-catenin and transcription factor CDX2.(6)Effects of Xianhecao on ethanol model rats tight junction proteins,adhesion junction proteins and transcription factors.Xianhecao could increase the expression of model rat tight junction proteins(Claudin-1、Occludin、ZO-1),adhesion junction proteins(E-cadherin、α-catenin)and transcription factors SOX2(P<0.05 compared with the model group).There was no effect on the expression of β-catenin transcription factors and CDX2.ConclusionsWuzhimaotao can prevent and treat gastric mucosal injury induced by indomethacin.Xianhecao can prevent and treat gastric mucosal injury induced by ethanol.The mechanism is associated with the improve expression of tight junction proteins,adhesion junction proteins and SOX2.It provides a reference for discussing the mechanism of prevent gastrointestinal mucosal injury of Wuzhimaotao and Xianhecao.
