Objective:To observe the effects of Electroacupuncture with different frequencies on intestinal transit function,colonic electromyography,nitric oxide synthase(NOS)content and interstitial cells of Cajal(ICC)in rats with slow transit constipation(STC).Methods:50 healthy male Wistar rats were randomly selected as normal group,and the rest 40 rats were fed with compound phenethylpiperidine,the dose of which was 8mg·kg-1·d-1,and were continuously administered for 120 days.40 rats were successfully established STC rat model,and randomly divided into model group,low-frequency electroacupuncture group,high-frequency electroacupuncture group,change group Frequency electroacupuncture group,10 in each group.The normal group and the model group did not receive any treatment,the other groups were given 2Hz continuous wave electroacupuncture(low frequency electroacupuncture group),100Hz continuous wave electroacupuncture(high frequency electroacupuncture group),2Hz/100Hz dense wave electroacupuncture(frequency conversion electroacupuncture group),the current intensity was about 2.0 Ma,once a day,for 15 minutes,a total of 15days.After treatment,the intestinal transport function,colonic electromyography,NOS content and the area of c-kit positive cells were measured in each group.Results:compared with the normal group,the excretion time of the first black stool in the other groups was significantly longer(all P<0.05);compared with the model group,the excretion time of the first black stool in the STC group was significantly shorter(all P<0.05)in the low frequency,high frequency and variable frequency electroacupuncture groups;compared with the low frequency electroacupuncture group and high frequency electroacupuncture group,the excretion time of the first black stool in the variable frequency electroacupuncture group was significantly shorter(all P<0.05);There was no significant difference between low frequency electroacupuncture group and high frequency electroacupuncture group(P>0.05).Compared with the normal group,the amplitude of colonic electromyography in the other groups was significantly larger and the frequency was faster(all P<0.05);compared with the model group,the amplitude of colonic electromyography in the three electroacupuncture groups was significantly smaller and the frequency was slower(all P<0.05);compared with the low frequency electroacupuncture group and the high frequency electroacupuncture group,the amplitude and the frequency of colonic electromyography in the frequency conversion electroacupuncture group were significantly smaller(all P<0.05);there was no significant difference between low frequency electroacupuncture group and high frequency electroacupuncture group(P>0.05).Compared with normal group,the content of NOS in other groups increased significantly(all P<0.05);compared with model group,the content of NOS in three electroacupuncture groups decreased significantly(all P<0.05);compared with low frequency electroacupuncture group and high frequency electroacupuncture group,the content of NOS in frequency conversion electroacupuncture group decreased significantly(all P<0.05);compared with high frequency electroacupuncture group,low frequency electroacupuncture group had no statistical significance(P>0.05).Compared with the normal group,the area of c-kit positive cells(ICC)in other groups decreased significantly(all P<0.05);compared with the model group,the area of c-kit positive cells in the three electroacupuncture groups increased significantly(all P<0.05);compared with the low-frequency electroacupuncture group,the area of c-kit positive cells in the frequency conversion electroacupuncture group increased significantly(P<0.05);compared with the high-frequency electroacupuncture group,there was no significant difference between the two groups(all P>0.05).Conclusion:The intestinal function of STC model rats can be improved by regulating the level of colonic myoelectric,NOS content and ICC expression.The effect of 2 Hz/100Hz frequency conversion therapy is better than that of 2Hz and 100Hz.
