Objective By detecting the relevant indicators in the liver tissue of each group of hepatic fibrosis rats and the expression levels of Vimentin,E-cadherin and β-catenin gene m RNA and protein,to clarify the mechanism of Yangying Huoxue Decoction against liver fibrosis and enrich the liver fiber Dissolve the pathogenesis connotation of "Ying deficiency and blood stasis".Methods 60 SD rats were randomly divided into 5 groups: normal control group,model control group,Yangying Huoxue Decoction treatment group,Fufang Biejia Ruangan Tablet treatment group and colchicine treatment group,with 12 rats in each group.The model was created by creating a model of liver fibrosis induced by carbon tetrachloride(CCL4).The paraffin oil and 40% CCL4 mixture and pig serum were alternately injected every 2 days.After 8 weeks,it was judged whether the establishment was successful.A rat liver fibrosis model was used to determine whether to continue the injection.To judge whether the model is successfully established,observe the symptoms and signs and make pathological sections.After the model was successfully established,each treatment group was given the corresponding drugs at the specified time every day for intragastric administration.At the same time,the normal control group and the model control group were given the corresponding amount of physiological saline solution to observe and record the basic situation of the moving rats.After 8 weeks of administration,liver tissue samples were collected and Real-time PCR and Western blotting were used to detect and analyze theIVexpression of Vimentin,E-cadherin and β-catenin in the liver tissues of rats in each group,and monitor their expression levels.Results 1.Compared with the normal control group,the model control group,Yangying Huoxue Decoction treatment group,compound Biejia Ruangan tablet treatment group and colchicine treatment group vimentin and β-catenin m RNA and protein expression levels were higher than those of the normal control group(P<0.05);Compared with the model control group,the m RNA and protein expression levels of vimentin and β-catenin in the Yangying Huoxue Decoction group,the compound Biejia Ruangan tablet group and the colchicine treatment group were lower than the model control group Group(P<0.05);Compared with the Yangying Huoxue Decoction treatment group,there was no significant difference in the m RNA and protein expression levels of vimentin and β-catenin in the Compound Biejia Ruangan Tablet treatment group(P>0.05);Compared with the colchicine treatment group For comparison,the m RNA and protein expression levels of vimentin and β-catenin in the Yangying Huoxue Decoction treatment group were lower than those in the colchicine treatment group,and there was a significant difference(P<0.05).2.Compared with the normal control group,the model control group,the Yangying Huoxue Decoction treatment group,the compound Biejia Ruangan tablet treatment group and the colchicine treatment group E-cadherin protein expression and m RNA transcription levels were lower than the normal control group(P<0.05);Compared with the model control group,the Yangying Huoxue Decoction group treatment group,the compound Biejia Ruangan tablet treatment group and the colchicine treatment group E-cadherin protein expression and m RNA transcription level were higher than the model control group(P <0.05);Compared with the Yangying Huoxue Decoction treatment group,the E-cadherin protein expression and m RNA transcription level in the compound Biejia Ruangan Tablet treatment group were not significantly different(P>0.05);compared with the colchicine treatment group,Yangying The E-cadherin protein expression and m RNA transcription levels of the Huoxuetang treatment group were higher than those of the colchicine treatment group,and there was a significant difference(P<0.05).3.Compared with the model control group,the m RNA and protein expressions of vimentin and β-catenin in the Yangying Huoxue Decoction group,the compound Biejia Ruangan tablet treatment group,and the colchicine treatment group were significantly reduced(P<0.05),but,The m RNA transcription level and protein expression of E-cadherin gene were higher than the model control group(P<0.05).3.The pathological results showed that the tissues of the model control group showed degeneration and necrosis,and inflammatory cells infiltrated into the portal area and central vein.The liver cells even showed vacuolar changes,pseudolobules appeared,and some fibrous tissues were abnormal.Hyperplasia,this is the overall pathological state of the model control group.Compared with the normal control group,the fibrosis is more obvious;compared with the model control group,the abnormal manifestations of liver cells in each treatment group are alleviated,and the fibrous tissue proliferation is also less.4.The values of serum liver fibrosis markers HA,LN,IV-C,PCⅢ and serum ALT and AST levels in the colchicine treatment group,Yangying Huoxue Decoction treatment group and compound Biejia Ruangan Tablet treatment group were lower than the model The control group(P<0.01),but higher than the normal control group(P<0.05).Conclusion Yangying Huoxue Decoction can reduce the related indicators in the serum of rats with liver fibrosis,by inhibiting the relative amount of vimentin and β-catenin gene m RNA transcription level and the expression of vimentin and β-catenin protein,promoting the E-cadherin m RNA transcription level and the expression of protein can achieve the effect of alleviating liver fibrosis.
