Study On The Anti-inflammation-cancer Activity And Material Basis Of A. Unilateralis

"Inflammation"and"cancer"of the two words on the current public is no longer strange.Inflammation refers to the response when bodies'tissue and cells are attacked;and cancer is a general term for all malignant tumors.Recently,many studies have pointed out that some of the tumor accompany with the chronic inflammation during the stage of occurrence and development,which implies that the malignant transformation of chronic inflammation is a key role in the progress of cancer.Zanthoxylum dissitum Hemsl.belongs to Zanthoxylum,it is spicy,bitter,and warm,to the liver,stomach.Although it is not recorded into Chinese Pharmacopoeia and has long been regarded as a fake of Zanthoxylumnitidum,it has a long history of medication in the folk.Zanthoxylum dissitum Hemsl.has a great value of promotion and clinical medicinal value,for example,in national Chinese medicine protective compound,Fuke Qianjin Tablets,Zanthoxylum dissitum Hemsl.plays anti-inflammatory effect.However,there are few studies on Zanthoxylum dissitum Hemsl.for"inflammation-cancer".Therefore,my experiment is based on the literature review and preliminary research of our group.Selected the stem of Zanthoxylum dissitum Hemsl.as the study object,using 70%ethanol reflux extracted then get total extract and successively extracted with CHCl₃,Et OAc,and n-Bu OH,then obtained three polar areas and determined the total alkaloid content in the total extract;The antimicrobial activity of total alkaloids and polar parts of single needle was determined by micro-turbidimetry;established the inflammatory model of macrophage RAW264.7 and the three kinds of cancer cells of hepatocellular carcinoma Hep G2,breast cancer MCF-7 and gastric cancer BGC-803 as the research object,try to explore the material basis of“inflammation-cancer”;Using conventional column chromatography and purification were carried out to separate the compounds from the stem of Zanthoxylum dissitum Hemsl.,and 20 compounds were identified by nuclear magnetic resonance.The results are as follows.1.The antimicrobial activity of total alkaloids and polar parts of Zanthoxylum dissitum Hemsl.was determined by microporous turbidimetric method.The results showed that all the samples had good antibacterial activity against the experimental strains in this experiment.The antibacterial effect of Gram-positive bacteria is better.2.The cytotoxicity of the whole alkaloid,polar parts and some monomeric compounds on the mouse macrophage RAW264.7 was detected by CCK8 method.The results showed that the concentration of macrophages in RAW264.7 was significantly higher than that in the control group Cells were not cytotoxic.3.Using Griess method to test the production of NO treat with total alkaloid,polar parts and some monomeric compounds from stem of Zanthoxylum dissitum Hemsl..The results showed that all the sample are showed a certain inhibition and the total alkaloid,CHCl₃ extract,nitidine chloride and dictamnine are Presented better activity.4.The cytotoxicities to Hep G2,MCF-7 and BGC-803 of all samples from the stem of Zanthoxylum dissitum Hemsl.was determined by the method of CCK8.The results suggested all of the samples showed positive activities and the anti-proliferative effect of BGC-803 on gastric cancer cells was the best in all the experimental groups.5.The chemical constituents of the polar parts of the single needle were analyzed by column chromatography and other methods.The structures of the 20 compounds were confirmed by NMR and mass spectrometry.They were:?-sitosterol 1,physcin2,dictamnine 3,liriodenine 4,?-sitosenone 5,vanillic acid 6,stigmasteryl-3,6-dione7,aurantio-obtusin 8,?-daucosterol 9,skimmianine 10,haplopine 11,scopoletin 12,isoscopoletin 13,4-methoxy-1-methyl-2-quinolone 14,nitidine chloride 15,nicotinic acid 16,glucosyringic acid 17,erythritol 18,uracil 19.Among then,compounds 2,5?8,12?13 and 16?19 are isolated from the plant for the first time.Additionally,compounds2,7,8,17and 20 are firstly isolated from the genus Zanthoxylum as well as the Rutaceae family,among then compound 20 is new.In this study,we chose to explore the related substances and theoretical basis of anti-inflammatory and anti-cancer activities from the aspects of antibacterial,anti-inflammatory and anti-cancer.The results of this experiment will be the follow-up research and development of Zanthoxylum dissitum Hemsl.provides a theoretical basis.
