Objective:Intervention of helicobacter pylori associated gastritis mice with spleen-stomach damp-heat sy-drome by Mieyou decoction of clearing heat and removing dampness method to observe the protein expression of PTEN,PI3 K,p-Akt and FoxO3 a and gene content of PTEN,PI3 K,Akt and FoxO3 a to explore the possible mechanism of Mieyou decoction in treating heli-cobacter pylori associated gastritis mice with spleen-stomach damp-heat syndrome.Methods:(1)60 BALB/c mice were randomly divided into 6 group,compound etiology model group,high-dose Mieyou Decoction group,medium-dose Mieyou Decoction group,low-dose Mieyou Decoction group and stomach quadruple group.After successful modeling,mice in control group and model group were given distilled water by gastric lavage,and mice in her group were given corresponding drugs by gastric lavage for 14 days respectively.(2)Index detection: HE staining was used to observe pathological changes and inflammation degree of gastric mucosa;TUNEL method was used to detect apoptotic rate of gastric mucosa;Western-blot was used to detect PTEN,PI3 K,p-Akt,FoxO3 a protein;RT-PCR was used to detect the expression of PTEN,PI3 K,Akt and FoxO3 a protein.Results: The expression and gene content of PTEN and FoxO3 a in model group increased,the expression of PI3 K,p-Akt and PI3 K,Akt gene content decreased,compared with the normal control group,the difference was significant(P<0.01);the expression of PTEN and FoxO3 a in high-dose Mieyou Decoction group,middle-dose Mieyou Decoction group,stomach quadruple group decreased,the expression of PI3 K,p-Akt protein and the content of PI3 K and Akt gene.Compared with the model group,the expression of PTEN and FoxO3 a protein and their gene content in the low-dose Mie Decoction group decreased,the expression of PI3 K and p-Akt protein and the content of PI3 K and Akt gene increased,but there was no significant difference between the two groups(P > 0.05).The expression of PTEN and FoxO3 a protein and their gene content decreased,the expression of PI3 K and p-Akt protein and the content of PI3 K and Akt gene increased in the high-dose Mie Decoction group,but there was no significant difference between the middle-dose Mieyou Decoction group and the middle-dose Mie Decoction group(P > 0.05).Conclusion : The high and middle doses of Mieyou decoction may inhibit HP-induced apoptosis of gastric mucosa cells through PTEN-PI3K-Akt-FoxO signaling pathway and play a role in treating HAG with spleen-stomach dampness-heat syndrome. |
