Effect Of Qiqi Wendan Decoction On Hsp60 In Gastric Mucosa And Serum IL-8 Of H.pylori-related Gastritis (Spleen Deficiency Damp-heat Syndrome) Model Mice

Objective: To observe the effect of Qilian Wendan Decoction on the levels of Hsp60 in gastric mucosa and serum IL-8 of Helicobacter pylori-associated gastritis(Spleen-damp-heat syndrome),and to explore the effect of Qilian Wendan decoction on Helicobacter pylori-associated gastritis Spleen deficiency damp heat syndrome)mechanism for the treatment of Helicobacter pylori associated gastritis to provide a new method.Methods: 60 Bal B / c mice with grade 6-8 weeks of SPF were divided into 6 groups,ie,model group,and the weight of male group was 20 ± 147 g,female: male = 1: 1.The rats in the middle,low dose group,western medicine(Livzonwei triple)group and blank control group were used.In addition to the blank control group,the other five groups were using composite factors(Hp tonic infection + simulated hot and humid environment + high fat and high sugar diet)to establish H.pylori-related gastritis spleen deficiency syndrome syndrome,after modeling success,the The rats in the model group and the control group were treated with the same amount of normal saline,and the rats in the control group were treated with the same amount of normal saline for 1 day / day for 14 days.The expression of HSP60 protein in the gastric mucosa of mice was detected by Western blotting.The expression of IL-8 in the serum of mice was detected by ELISA.Results:(1)The mental state,coat color,activity amount,food intake,stool and so on were all improved in the model group.(2)H.pylori colonization and pathological damage of gastric mucosa were observed under light microscope.Compared with the blank group,the H.pylori colonization and pathological score of the model group were higher than those in the blank group(P <0.01).Compared with the model group,the dose group and the Lizhuwei triple The H.pylori colonization and pathologic score were lower in the model group than in the model group(P <0.05).(3)Compared with the blank group,the expression of Hsp60 and serum IL-8 in the gastric mucosa of the model group were higher than those in the blank group(P <0.01).Compared with the model group,The expression of Hsp60 and IL-8 in gastric mucosa were lower than that in model group,p <0.05.Conclusion:(1)self-made Qilian Wendan Decoction can improve H.pylori-related gastritis(Spleen damp-heat syndrome)mice in general,reduce H.pylori colonization,improve H.pylori-related gastritis(Spleen damp-heat syndrome)gastric mucosal pathology On H.pylori associated gastritis(spleen damp heat syndrome)have a better therapeutic effect.(2)It's mechanism may be by reducing the expression of HSP60 in the gastric mucosa,reducing the anti-HSP60 antibody produced by Helicobacter pylori and the corresponding part of the corresponding cross-reaction,inhibition of antigen and antibody cross reaction;reduce serum IL-8 expression,inhibition of inflammation Cell release,reduce inflammation damage,so as to achieve therapeutic effect.
