| Grape seeds are rich in nutritional components,particularly polyphenols.In the present work,with water as the solvent,the extraction technology of polypheol from grape seeds was optimized,and several measures were taken to improve the stability and improving the taste of the solvents.In addition,the oral liquid qualitative standards were proposed.The results were as follows:(1)optimal operation conditions were found through orthogonal tests:grape seeds were grounded into particles with which size were 20 mesh;the samples were then dipped in hot water(95℃)for 3 hours,with the ratio of sample and solvent was 1:9.Repeating the operation for one time,and the concentration of polypheols was about 1g/L.This extraction was kept at 0-4℃ for 48hours,and after adapting its pH to about 4.0,the solvent was separated from grinding particles.(2)the stability test showed:the concentration of polyphenols were significantly influenced by using of gelatin.An appreciable reduction was shown when 0.01%gelatin was used.However,bentonite had little effect on polyphenols.After treated by 0.3%bentonite the extraction would be clarified.(3)the result of blending test:when the extraction was mixed with sucrose(6%)and citric acid(0.15%),most people could handle its taste.Meanwhile,benzoic acid(100mg/L)and sorbic acid(400mg/L)were used in case of the extraction was rotted.(4)microbial stability test’s result:the extraction was heated by 60℃ for 30minutes,85℃ for 15minutes and 95℃ for 5 seconds respectively,but these three type measures all had little effect on the concentration of polyphenols.(5)Since there is no related standards about polyphenols oral liquidfrom grape seeds or similar products.Here,we developed some qualitative standards of this oral liquidaccording to the contracts on the oral liquid in China Phannacopeia2000,first section.The qualitative standards included:this oral liquid was light yellow to reddish-brown,Crystal-clear and had the special flavor of grape seeds,with the concentration of polyphenols in this oral liquidwas not less than 1g/L and the total solids content was no less than 6%. |
