| Sesame lignan has unique and excellent nutritional and physiological activities,including free fat soluble lignan and water soluble lignan containing glycosome.In recent years,there are relatively many studies on sesamin,but few studies on sesaminol and glucoside products.However,sesaminol and its glucosidylated products may have better biological activity,and the products are more likely to play a role in intestinal tract.Therefore,this paper through the intestinal flora in vitro cultivation of anaerobic fermentation and cell culture,in vivo animal studies sesaminol and its glucosidylated products structure of intestinal flora,intestinal probiotics proliferation and adhesion of the adjustment and promote role,clarify the sesaminol and its glucosidylated products products improve the intestinal microecology,promotes intestinal health of biological activity and structure function relationship.The results are as follows:1.In vitro fecal anaerobic fermentation system was used to investigate the effects of sesaminol and its glucosidylated products on intestinal microflora of normal diet mice by high-throughput sequencing technology.It can be seen that sesaminol and sesaminol glucoside could significantly increase the abundance of intestinal probiotics Lactobacillus and Bifidobacteria,and reduce the abundance of Escherichia coli.Both sesaminol and its glucosidylated products can regulate the structure of intestinal flora and the diversity of intestinal microbes,and significantly increase the relative abundance of Enterococcus and Lactobacillus.The relative abundance of Parabacteroides,Bacteroides and Eubacterium was decreased.It can also significantly increase the content of short-chain fatty acids and reduce the intestinal p H value.Therefore,in the anaerobic fermentation experiment of intestinal flora in vitro,both sesaminol and its glucosidylated products can regulate the structure of intestinal flora,promote the proliferation of intestinal probiotics and improve the intestinal microecological environment in vitro,and the regulation effect of sesaminol and its glucosidylated products on intestinal flora is similar.2.In vitro cell culture experiment was conducted to investigate the effects of sesaminol and its glucosidylated products on the adhesion of intestinal probiotics to colon epithelial cells(NCM460).The results showed that both sesaminol and sesaminol glucoside could promote the adhesion of probiotics(Bifidobacterium bifidum and Lactobacillus acidophilus)to NCM460 cells.Moreover,sesaminol and sesaminol glucoside can up-regulated the gene and protein expression levels of adhesive proteins(β-catenin,E-cadherin)in NCM460 cells.Therefore,in vitro cell culture experiments,both sesaminol and its glucosidylated products can promote the adhesion of intestinal probiotics and the expression of adhesive proteins in colon epithelial cells,the effect of sesaminol and its glucosidylated products on the adhesion of intestinal probiotics is similar.3.C57BL/6J mice were fed a normal diet supplemented with probiotics to investigate the effects of sesaminol and its glucosidylated products on intestinal microflora structure,adhesion of intestinal probiotics and intestinal microecology.The results showed that both sesaminol and sesaminol glucoside could significantly increase the abundance of probiotics Lactobacillus and Bifidobacterium,and decrease the abundance of Enterococcus and Escherichia coli in intestinal contents of mice,decrease p H value of cecal contents and increase short chain fatty acid content.Sesaminol and its glucosidylated products could change the structure and species of intestinal flora,increase the relative abundance of Lactobacillus,Bifidobacterium and Akkermansia,and decrease the relative abundance of Allobaculum and Bacteroides.Compared with sesaminol,sesaminol glucoside could significantly increase the relative abundance of intestinal probiotics including Bifidobacteria and Akkermansia,and significantly increase the intestinal microbial diversity.In addition,sesaminol and sesaminol glucoside significantly up-regulated the gene and protein expression levels of adhesive proteins(β-catenin,E-cadherin)and tight junction proteins(Occludin,ZO-1)in colon tissues of mice.Moreover,compared with sesaminol,sesaminol glucoside significantly increased the gene and protein expression levels of adhesive proteins(β-catenin,E-cadherin)and tight connexin(ZO-1)in mice colon tissues.Therefore,sesaminol and its glucosidylated products can regulate the structure diversity of intestinal flora,promote proliferation and adhesion colonization of intestinal probiotics,and protect the integrity of intestinal barrier function.Compared with sesaminol,the effect of sesaminol glucosidylatated products was more significant,indicating that the structure of sesaminol glucosidylation can better play the intestinal probiotics activity. |
