| Ethnic traditional dairy food is a dairy food processed and made by Mongolian traditional pure milk raw materials,traditional crafts,and traditional production and processing methods,which is collectively known as national traditional dairy milk food.There are 7 varieties of traditional national dairy products that have formulated local food safety standards in the autonomous region,namely: Bishilag,Zege,Milk Skin,Chula,Milk Tofu,Sour Cheese Egg and Chewing Gram.The purpose of this paper is to establish a method for the simultaneous detection of benzoic acid,sorbic acid and dehydroacetic acid in ethnic traditional dairy foods by high performance liquid chromatography.It is through experimentation,finding the best chromatographic conditions and simple and fast sample pretreatment methods,so as to achieve a simplified process of sample handling operations.Due to the different configurations of the laboratory instruments,this test determines the high performance liquid chromatography conditions suitable for the state of the laboratory instruments through exploration,and the laboratory configuration is Thermo Fisher Scientific high performance liquid chromatography,diode array detector(DVD),C18 column.Through the scanning of the ultraviolet absorption spectrum of the above three food additives,the ultraviolet detection wavelength of this test was determined to be 230 nm;By consulting the literature,the above three food additives are mostly detected using methanol,ammonium acetate solution(0.002mol/L)system,and the mobile phase of this test adopts methanol + ammonium acetate solution(0.002mol/L)= 5 + 95,the flow rate is 1m L / min and other gradient elution,column temperature 30 °C.The sample pretreatment method was compared through multiple tests to determine the best extraction and purification method.Under the above liquid chromatography conditions and sample pretreatment operations,the method was confirmed that the linear correlation coefficient of the three kinds of food additives in the range of 0-40ug/m L was better than 0.999;the detection limit3 times signal-to-noise ratio calculation and quantitative limit 10 times the signal-to-noise ratio calculation were less than the national standard requirements and the sensitivity was high;Precision is expressed in RSD values,all less than 5%;The accuracy is expressed in terms of recovery rate,which is between 90% and 110%,and all meet the requirements of GB/T 27417-2017.The test results prove that the method is effective and feasible,and can be used for actual production,saving the cost of detection and improving the efficiency of detection. |
