Analysis On The Difference Of Metabonomic And Physiological Of Three Gentiana Plants | | Posted on:2024-05-10 | Degree:Master | Type:Thesis | | Country:China | Candidate:W Y Wu | Full Text:PDF | | GTID:2543306932489154 | Subject:Forest science | | Abstract/Summary: | PDF Full Text Request | | Radix Gentianae,as a traditional Chinese herbal medicine in northeast China,is derived from the dried roots and rhizomes of Gentiana manshurica,Gentiana triflora,Gentiana scabra or Gentiana rigescens,belonging to Gentianaceae.The first three species are known as Gentiana scabra,the main producing area is in the northeast,and the latter species is known as Gentiana rigescens,and the main producing area is in Yunnan.In order to explore the metabolic differences of medicinal parts of three Gentiana plants in Northeast China,to explore the metabolic differences of aerial parts(leaves and flowers)of three Gentiana plants and their medicinal parts,and to explore the metabolic differences of medicinal parts of Gentiana scabra in different environments,this study used three Gentiana plants as materials,Through the qualitative and quantitative detection of GC-MS non-targeted metabonomics technology platform and the comprehensive analysis of the results of physiological and biochemical indicators,the results are as follows:(1)The metabolic differences of root tissues of medicinal parts of three Gentiana plants were discussed.The results of PCA and PLS-DA analysis showed that there were differences in root tissues of three Gentiana plants.Fifty-eight kinds of differential metabolites were screened from the root tissues of three Gentiana plants,among which most of the differential metabolites such as L-Glutamic acid,L-Alanine and L-Prolind were accumulated in the root tissues of Gentiana scabra,and all of them participated in the ABC transporter pathway,which was related to the response of plants to external stress and the transport of secondary metabolites.At the same time,the above amino acids can be directly used as intermediates for medical supplies or synthetic medicines.The results showed that Gentiana scabra may be better than Gentiana triflora and Gentiana manshurica in coping with external stress and accumulation of secondary metabolites and amino acids,which provided data basis for the identification of Radix Gentianae.(2)The metabolic differences in different tissue parts of three Gentiana plants were discussed respectively.The results of PCA and PLS-DA showed that there were differences among different tissue parts of three Gentiana plants.49,50 and 55 different metabolites were screened from different tissue parts of Gentiana manshurica,Gentiana triflora and Gentiana scabra,respectively.Cluster analysis showed that the contents of some sugar and acid metabolites in leaves and flowers of three Gentiana plants were higher than those in roots.Metabolic pathway analysis shows that there are differences in the metabolism of different tissue parts of three Gentiana plants,and this difference is less affected by Gentiana plants.Although all three Gentiana plants are widely used as medicinal parts,the research results show that the contents of some metabolites in leaves and flowers of three Gentiana plants are higher than those in roots,which provides data support for the development and utilization of aboveground parts of three Gentiana plants.(3)The metabolic differences of Gentiana scabra in different growth environments were discussed.The results of PCA and OPLS-DA showed that there were differences in the metabolism of Gentiana scabra samples grown in different environments,and 55 different metabolites were screened between the two groups.The accumulation of most of the differential metabolites is high in the root tissue of Gentiana scabra in wild environment.The metabolic pathway analysis shows that the differential metabolites participate in amino acid metabolism,sugar metabolism,organic acid metabolism and phytohormone synthesis,which indicates that different growth environments may affect the accumulation of amino acids,sugars,organic acids and phytohormones in Gentiana scabra.(4)The analysis results of soluble sugar and soluble protein showed that the accumulation of soluble sugar and soluble protein in root tissue of Gentiana scabra was higher than that of other two Gentiana plants.Environment may be the reason for the difference of soluble sugar and soluble protein accumulation in root tissue of Gentiana scabra,and wild environment may be more conducive to the accumulation of soluble sugar and soluble protein in root tissue of Gentiana scabra.The above-ground parts of three Gentiana plants may also have beneficial substances that can be developed and utilized. | | Keywords/Search Tags: | Gentiana manshurica, Gentiana triflora, Gentiana scabra, Metabonomic, GC-MS | PDF Full Text Request | Related items |
