| Zanthoxylum bungeanum(Zanthoxylum bungeanum Maxim.)is a deciduous small tree of the genus Zanthoxylum(Zanthoxylum L.)in the family Rutaceae(Brassicaceae).Zanthoxylum bungeanum has excellent quality,strong aroma and full fruit,and is widely distributed in the Yanshan Mountains,However,the variety identification has not been conducted for the time being.At present,the propagation of Yanshan pepper is mainly live propagation.The uneven quality,low yield and more serious pests and diseases become a problem.Use the shoot cuttings can maintain the excellent traits of the parent while also allowing for the study of prickle degeneration and early fruiting of pepper.In this thesis,different exogenous hormones and concentration gradients were used to study the propagation of shoot cuttings of Zanthoxylum bungeanum.At the same time,nalysed the rooting of cuttings,the changes of nutrient composition in the base of cuttings and variation trend of enzyme activities of different kinds after cutting.The conclusion is as follows:1.The occurrence of adventitious roots in shoot cuttings of Zanthoxylum bungeanum is of mixed rooting type.The process of shoot cuttings of Zanthoxylum bungeanum was divided into four periods: 0-10 d after shoot cuttings for the induction period of pepper guaiac tissue;20-30 d after shoot cuttings for adventitious root formation;30-40 d after shoot cuttings for adventitious root elongation;40 d after shoot cuttings for late rooting.2.From the experiments with different exogenous hormones and their concentrations,it was found that NAA treatment was more effective than IBA treatment in the single hormone treatment,among which NAA treatment at 400 mg/L was the best,with a rooting rate of 33.33%.Among the mixed hormone treatments,500 mg/L mixed ratio of IBA and NAA(1:3)had the best effect with a rooting rate of76.67%.3.Selection of the lower cut of the spike: The selection of the lower cut site can affect the rooting of pepper spike,the rooting rate of the diagonal cut at the node was60.00%,and the rooting rate of the diagonal cut between nodes was 33.33%.4.In the rooting process of Zanthoxylum bungeanum shoot cuttings,two enzymes,POD and SOD,were found to be closely related to the rooting of shoot cuttings.The high activity of SOD can make the cuttings heal quickly and form healing tissue,thus shortening the growth time of adventitious roots;the increase of POD activity is beneficial to the formation of adventitious roots of the cuttings. |
