| Sow production performance is an important indicator used to measure the benefits of modern intensive pig farming practices.However,dramatic changes occur in perinatal sows,resulting in low immunity.The process of sow production is affected by many factors such as feeding management,nutrition supply and health status.It is prone to a series of problems such as fewer piglets,long labor,insufficient lactation and low survival rate of weaned piglets.The above problems seriously reduce the production performance of sows and affect the economic benefits of pig farms.Based on the theory of traditional Chinese veterinary medicine,the research group improved the classic Chinese medicine prescription Bazhen decoction.Through clinical trials,it was proved that the addition of modified Bazhen powder during perinatal period could improve the production performance and immunity of sows.However,in the previous experiment,the modified Bazhen powder was added in the form of ultrafine powder,which was prone to uneven mixing and poor palatability.Therefore,the development of new dosage forms that are more suitable for clinical application is of great significance in order to efficiently apply the modified Bazhen powder.The dry suspension is used in drinking water,and the palatability is good.At the same time,Chinese medicine is uniformly dispersed in water in the form of particles and can be absorbed quickly.Aiming at the above problems,this study prepared the modified Bazhen powder dry suspension,and optimized its prescription process.After clinical verification,the preparation can improve the production performance and immune function of sows during the perinatal period.The results in this study are as follows:(1)The optimal suspending agent,wetting agent and flavoring agent were selected by single factor test,which were xanthan gum,poloxamer-188 and sucrose.The settlement volume ratio and re-disperse capability were used as the test indexes.The prescription of dry suspension was optimized by orthogonal design test.The optimal prescription is: each 1 g modified Bazhen powder dry suspension contains the main drug 0.32 g,xanthan gum 0.072 g,poloxamer 188 0.06 g,sucrose0.548 g.(2)Check the modified Bazhen powder dry suspension according to the general rules of preparation.The modified Bazhen powder dry suspension is light yellow powder with a rough microscopic surface.The particle size is between 4-6 μm and the absolute value of the potential is between 39.1-44.2 m V.The sedimentation volume ratio is greater than 0.9,the re-disperse capability is good,and the drying weight loss is less than 2 %.The stability test showed that the preparation was stable to temperature and light.However,it is greatly affected by humidity,so high humidity environment should be avoided during storage.The preparation can be stored at temperature below40 °C and humidity below 75 % for at least 6 months.Quality inspection meets the requirements of pharmacopoeia.(3)The modified Bazhen powder dry suspension was administered by drinking water.The production performance and immune indexes of sows were determined in this experiment.The results showed that the modified Bazhen powder dry suspension could significantly increase the number of healthy offspring,reduce the number of weak offspring,shorten the labor process and the birth interval;significantly improve the birth weight of piglets,weaning litter weight,sow daily lactation;significantly increase the content of milk protein,milk fat and dry matter in colostrum;significantly increased serum follicle stimulating hormone,luteinizing hormone,estrogen content;and significantly increased serum immunoglobulin G,immunoglobulin A content.(4)The blood components of traditional Chinese medicine were determined by liquid chromatography-mass spectrometry.A total of 37 components such as liquiritin,glycyrrhiza coumarin,Gancaonin G,aloe-emodin,inositol,ent-17-hydroxy-16beta-kaura-19-al,phytolacca saponin A and yangonin were identified in sow serum.The above Chinese medicine into the blood components are mainly attributed to flavonoids,terpenoids,phenylpropanoids,etc.It provides a theoretical basis for elucidating the possible mechanism of action of Chinese medicine into blood components in sows.In summary,this study successfully prepared and optimized the prescription of modified Bazhen powder dry suspension by using single factor test and orthogonal test.The preparation method of the dry suspension is simple and suitable for large-scale production.At the same time,the dry suspension has a positive effect on improving the production performance and immune indexes of sows.In addition,37 blood components of traditional Chinese medicine were identified by liquid chromatography-mass spectrometry,which provided a theoretical basis for clarifying the efficacy.This study not only provides a new dosage form for the clinical application of modified Bazhen powder,but also provides reference and theoretical basis for its application in improving production performance and immune performance of sows. |
