661W cells were cultured and photodamaged 661W cells model was established in vitro.661W cells were continuously irradiated with 2600±100Lux visible light for 48h.The 661W cells were cultured in groups:661W cells in the Normal control group(Normal),661W cells in the Light damage group(Light)and 661W cells in the safflower yellow pigment intervention group(Light+).The optimal concentration of safflower yellow pigment for light damage group was 50μmol/L.The experimental results showed that:(1)Scanning electron microscopy of 661W cells in each group showed that the morphology of 661W cells in Light+group was between Normal group and Light group.(2)The cell viability(100%)of 661W cells in Normal group,Light group and Light+group was 87.78±9.088,27.16±6.930 and 57.87±6.278,respectively,by cck-8 method,and the difference was statistically significant(P<0.001).(3)The MDA content(umol/mg port)of 661W cells in Normal group,Light group and Light+group was 1.04±0.103,2.47±0.209 and 1.82±0.129,respectively,with statistical significance(P<0.0001).(4)The SOD(U/mg port)of 661W cells in Normal group,Light group and Light+group was 83.41±2.393,43.89±1.460 and 61.18±4.193,respectively,and the difference was statistically significant(P<0.0001).(5)Western blot detected Cyp1b1 protein and key factor protein of pyrogenic pathway in each group,and the results showed:The protein expressions of Cyp1b1 protein and key factors of pyroptosis pathway in 661W cells of Light group were higher than those of Normal group,while those of Cyp1b1 protein and key factors of pyroptosis pathway in 661W cells of Light+group were lower than those of Light group,with statistical significance(P<0.05).(6)RT-qPCR was used to detect Cyp1b1 mRNA and the mRNA of key factors of pyrodeath pathway,and the results showed:The mRNA contents of Cyp1b1 mRNA and key factors of pyroptosis pathway in 661W cells of Light group were higher than those of Normal group,while the mRNA contents of Cyp1b1 mRNA and key factors of pyroptosis pathway in 661W cells of Light+group were lower than those of Light group,with statistical significance(P<0.01).
