| Objective:To investigate whether curcumin,exercise,and curcumin combined with exercise can prevent adiponectin secretion decline induced by a high-fat diet in rats,we did this research and explore its possible mechanisms.Methods:40 different animals to adaptive feed after 1 week,randomly divided into 5 groups,each group of eight,divided into normal control group(group C 8),a high-fat diet group(group H only 8),a high-fat diet and curcumin groups(HC group 8),a high-fat diet and aerobic exercise group(group HE 8),high fat diet plus exercise and curcumin group(X group 8).Group C was fed with ordinary feed.Group H,HC,HE and X were fed with high-fat diet.Meanwhile,curcumin was prepared into a suspension with 0.5%sodium carboxymethyl cellulose and given to HC and X groups by gavage of curcumin suspension once a day for 6 weeks at a dose of 200mg/kg body weight.Groups C,H and HE were gavaged with 0.5%sodium carboxymethyl cellulose.The HE and X groups were given 75%VO2max with curcumin intravagation at a gradient of 5 degrees,once a day,60 minutes a time,6 times a week(Sunday rest)for 6 weeks.36 h after the last exercise,the body weight of the rats was measured,then the rats were anesthetized,blood was collected,and then the rats were killed and the peritestosterone fat was collected.Fasting blood glucose(FBG),glucose tolerance,serum adiponectin and lipids(TC,TG,LDL-C,HDL-C)were measured.The protein levels of PPARy and DsBA-L in rat adipose tissue were detected by Western blot.Results:①Compared with group C,the body weight,FBG,serum TC,TG and LDL levels of rats in group H were increased(P<0.05),the HDL-C and glucose tolerance were decreased,and the protein levels of PPARγ(P<0.05)and DsbA-L(P<0.01)and adiponectin in group H were decreased(P<0.05).②Compared with the H group,the body weight,FBG,serum TC,TG,LDL-C levels of the rats in the HC group were decreased,while the HDL-C and glucose tolerance were increased(P<0.05).Meanwhile,the protein levels of PPARy and DsBA-L in adipose tissue of the rats in the H group were increased,and the adiponectin level was also increased(P<0,05).③Compared with H group,the body weight,FBG,serum TC,TG,LDL-C,HDL-C and glucose tolerance of rats in HE group were decreased(P<0.05),but the PPARy protein and DsBA-L protein levels in adipose tissue of rats in HE group were decreased,and there was no significant difference between them and H group,and the adiponectin level was also not significantly different.(P>0.05).④Compared with group H,the body weight,FBG,serum TC,TG,LDL-C,HDL-C and glucose tolerance were decreased in group X(P<0.05).Meanwhile,the protein levels of PPARy and DsBA-L in adipose tissue were increased in group X,and the adiponectin level was also increased(P<0.05).Moreover,the intervention effect of TC,TG,LDL-C,HDL-C,FBG and glucose tolerance in X group was better than that in HE group and HC group,and the activation effect of up-regulation of adiponectin by PPARy-DsbA-L in X group was slightly better than that in HC group.Conclusion:Curcumin,exercise or curcumin combined with exercise intervention can effectively inhibit metabolic syndrome such as dyslipidemia,elevated fasting blood glucose and decreased glucose tolerance in rats with high fat diet.But curcumin can be raised PPAR gamma protein expression and PPAR gamma-DsbA-L pathways through promoting adiponectin protein expression in order to restrain high fat diet rats adiponectin secretion decreased,while movement is not through the pathway of promoting adiponectin protein expression,movement joint of curcumin can PPAR gamma-DsbA-L pathways through promoting adiponectin protein expression and inhibiting a high-fat diet rats adiponectin secretion decline. |
