Effect Of 32-week HIIT And Weight-bearing Training Intervention On ADPN-PPARα Pathway In Liver Of Different Period Aging Rats

Objective:To investigate the effects of 32-week HIIT and weight-bearing training on lipid metabolism by regulating the hepatic ADPN-PPARa signaling pathway in the different time period of aging.Methods:We randomly chose 10 out of 130 eight-month-old male SD rats as baseline group,after 1 week of adaptive feeding,rats were executed.Then randomly divided rest of the rats into a control(group C),a HIIT group(group H)and a weightbearing training group(group R),40 rats in each group.Group H took 6 loops of 3min 90%VO2max-3min 50%VO2max speed treadmill running,with 5min 70%VO2max speed at the beginning and the end of the training,46min/day in total.Training was carried out 3 days/week,last for 32 weeks and then every 4 weeks perform a maximum oxygen uptake test and adjust the training speed according to the results.Group R was tested with the maximum weight-bearing capacity,and 30%of the maximum weightbearing capacity was used to climb on a treadmill with a slope of 35° and a running speed of 15m/min for 46 minutes/day,and training was carried out 3 days/week,for 32 weeks.After that,the maximum weight-bearing test was performed every 4 weeks and the weight-bearing weight is adjusted according to the results.24 hours after the end of every 8 weeks of intervention,10 rats were randomly chosen from each group,electronic weight meter was used to test the weight,DXA was used to test the body fat of rats in each group.Afterwards,samples were taken,abdominal aortic blood was taken,serum was separated,and GPO-PAP and ACS-PAP methods were used to test the content of TG and NEFA,and ELISA assay was used to test the content of ADPN;observe the percentage of lipid droplet area of the oil red O stained liver with a microscope,use GPO-PAP and ACS-PAP methods to test the content of TG and NEFA,and use Western Blot to test the liver ADPN,AdipoR2,PPARa and ACOX1 levels.All index data using SPSS for ANOVA analysis of varianceResults:During the 32-week aging and intervention process:1.There were significantly differences between group H,R and group C of the body fat percentage and liver lipid droplet area while at same time point,and group H,R were lower than group C and H;2.There were significantly differences between group H,R and group C of the serum TG while at same time point,and group H and R ware lower than group C.And at the 32th week,there was significantly difference between group H and group C of the serum ADPN,and group H was higher;3.The liver TG and NEFA of group H and R were significantly lower than group C at the same time point;4.At the 8th and 16th week,there were significantly differences,respectively,between group H,R and group C of the liver PPARα while at same time point,and group H and,R were higher than group C.At the 8th week,there were significantly differences,respectively,between group H,R and group C of the liver ACOX1 while at same time point,and group H and,R were higher than group C,while at 32th week,group R was significantly lower than group C.Conclusions:1.Performing HIIT and weight-bearing training for no less than 8 weeks can effectively delay or improve the accumulation of fat in body and liver in different period of aging,and two exercises may have no difference in long period;2.Performing HIIT and weight-bearing training for no less than 16 weeks can effectively delay the increasement of serum and liver lipids,and weight-beraing training may have better effect in long period;3.32-week HIIT intervention can increase the serum ADPN content compared with weight-beraing training;4.32-week HIIT and weight-bearing training can increase liver ADPN-PPARα pathway related protein expression within a certain period of time,and improve lipid metabolism.The effect of HIIT was more prominemt at 8th week,while weight-bearing training was at 16th week.
