Objective:To compare the effects of aerobic exercise and/or curcumin intervention on the improvement of intestinal flora disorders caused by long-term highfat diet,and to provide data support for the regulation and improvement of intestinal flora disorders induced by high-fat diet.METHODS:Only 5 weeks of age 40 male SD rats,after 7 days adaptive breeding,using the method of random Numbers were carried out in accordance with the weight group of 40 rats,divided into normal meal+quiet group(group C),high fat diet group(group H)+quiet quiet,high-fat diet+curcumin+group(J),high fat diet+aerobic exercise group(group S)and a high-fat diet+curcumin+aerobic exercise group(U).Rats in each group were fed with free food and drinking water,and group C was fed with ordinary feed.Groups H,J,S and U were fed with high fat diet(60%high fat calorie ratio diet).J group,U 3 week according to previous research results of the experiments lavage of curcumin(0.5%CMC-Na as a suspension agent,the mixture of curcumin suspension liquid stored at 4℃refrigerator spare)dose of 200 mg/(kg·d),the volume of 5 mL/kg,0.5 h before every training and give the dose lavage,1 x per day,the rest of the group in 0.5%sodium carboxymethyl cellulose by gastric volume,such as gastric six weeks.Group S and Group U received aerobic exercise intervention from the 3rd week.The exercise program was as follows:Slope:5°;exercise intensity:70%VO2 Max;duration of each exercise:60min;frequency:once a day,6 days a week;duration of intervention:6 weeks;other groups did not receive any exercise intervention.Samples were collected 24 hours after the last aerobic exercise,weighed before sampling,anesthetized with ulatan solution and killed,blood and cecum contents were collected for measurement.The concentration of lipopolysaccharide(LPS)in serum samples was determined by double-antibody sandwiched enzyme linked immunosorbent assay(ELISA).In aseptic conditions under the removal of cecal contents to be measured;16S rRNA gene sequencing was performed on the cecal intestinal contents,and the OTUs,alpha-diversity,betadiversity and inter-group differences of intestinal flora were analyzed to detect the diversity of intestinal flora.Results:The 16S rRNA gene sequence of 25 samples showed that(1)serum LPS activity in group H was increased compared with that in group C,and the difference was very significant(P<0.01);Compared with group H,serum LPS activity was decreased in group J,and the difference was very significant(P<0.01),LPS decreased significantly in group S(P<0.05),the LPS of U group was decreased,and the difference was extremely significant(P<0.01).(2)Compared with group H,curcumin intervention,aerobic exercise combined with curcumin intervention increased the similarity of OUT composition between the rats on high fat diet and group C.(3)In alpha-diversity analysis,the Chao1,Shannon,and Simpson indices of rats in group H were abnormal compared with those in group C(P<0.01),intestinal microflora in group H was disturbed,and the richness and diversity of intestinal microflora in groups S and J were significantly changed compared with group H(P<0.05),compared with group U,there were no significant changes in the richness and diversity of intestinal microflora in groups S and J(P>0.05).(4)in the structure of intestinal flora analysis found that:compared with the group H,J,S group,U group F/B ratio in the rat intestinal flora changes,and varying degrees of decline,J,S group of rat intestinal flora in F/B ratio significantly reduced(P<0.05),and rat intestinal flora in group U F/B ratio(P>there was no significant change0.05);Compared with group U,the F/B ratio of intestinal flora in groups J and S had no significant change(P>0.05).(5)In beta-diversity analysis,the contribution rates of major "elements in the three principal coordinates were 54.71%,17.33%and 10.9%,respectively.(6)ANOSIM similarity analysis confirmed the efficacy of the intervention on intestinal flora(P<0.01).(7)LEFSE analysis found that there were 13,10,6,1 and 11 important biomarkers in group C,H,J,S and U,respectively.Conclusion:Single aerobic exercise or curcumin intervention can improve the diversity of intestinal microflora in rats fed high fat diet.The combination of the two was no better than the single intervention.
