| Objectives:To study the effects of ultra-low temperature and cold therapy on NLRP3 inflammasome and its related signaling pathways,and explore the molecular physiological mechanism of its inhibition of inflammation,so as to provide reference for subsequent studies.To provide practical examples for the application of ultra-low temperature and cold therapy in hockey projects,enrich the means of recovery after training in hockey projects,and provide experimental basis for promoting the application and promotion of ultra-low temperature and cold therapy in sports practice.Methods:Twenty-eight players of Tianjin Men’s hockey team were selected and divided into experimental group and control group.There was no significant difference in age,height,weight,training years and sports grade between the two groups.The experimental group received ultra-low temperature cryotherapy at-130℃for 180 s twice a week for 5 weeks,while the control group did not receive ultra-low temperature treatment.In addition,the experimental subjects will also receive 42 minutes of routine rehabilitation(27min of stretch relaxation and 15 min of foam axis relaxation),and the control subjects will receive 45 minutes of routine recovery(30min of stretch relaxation and 15 min of foam axis relaxation).SPSS26.0 software was used to analyze the experimental data.t test was used for data conforming to normal distribution,and non-parametric test was used for data not conforming to normal distribution.Spearman’s rank correlation coefficient was used to analyze the correlation between the expression levels of NLRP3 m RNA,NLRP3,IL-18 and testosterone and cortisol,and the level of significance was P<0.05.Results: Results of inflammatory reaction:(1)Before intervention,there were no significant differences in NLRP3 m RNA,IL-1βm RNA and IL-18 m RNA expression between the experimental group and the control group(P>0.05).After intervention,compared with before intervention,there were no significant differences in the expressions of NLRP3 m RNA,IL-1βm RNA and IL-18 m RNA in the experimental group(P>0.05),no significant differences in the expressions of NLRP3 m RNA and IL-1βm RNA in the control group(P>0.05),and the expression of IL-18 m RNA was significantly up-regulated(P<0.05).After the intervention,the expression of IL-18 m RNA in the experimental group was significantly lower than that in the control group(P<0.05),and the expression of NLRP3 m RNA and IL-1βm RNA were not significantly different(P>0.05).(2)Before intervention,the levels of serum proinflammatory cytokines NLRP3,Caspase-1,IL-18 and IL-1β were not significantly different between the two groups(P>0.05),nor were the levels of serum anti-inflammatory cytokines IL-10 and IL-6(P>0.05);After intervention,compared with before intervention,the contents of pro-inflammatory cytokines NLRP3,Caspase-1,IL-1β and IL-18 in experimental group were significantly increased(P<0.01),the level of anti-inflammatory cytokines IL-6 was significantly increased(P<0.01),and the level of IL-10 was significantly decreased(P<0.01).In control group,the contents of pro-inflammatory cytokines NLRP3,Caspase-1,IL-1β and IL-18 were significantly increased(P<0.01),the content of anti-inflammatory cytokines IL-6 was significantly increased(P<0.01),and the content of IL-10 was significantly decreased(P<0.01);After intervention,the contents of proinflammatory cytokines NLRP3,IL-1β and IL-18 in the experimental group were significantly lower than those in the control group(P<0.05),while the levels of proinflammatory cytokines Caspase-1 and anti-inflammatory cytokines IL-10 and IL-6 were not significantly different(P>0.05).Results of functional state:(1)Before intervention,there were no significant differences in testosterone,cortisol,T/C value and CK content between the two groups(P>0.05).After intervention,compared with before intervention,the testosterone content in the experimental group was significantly increased,the cortisol level was significantly decreased,and the T/C value was significantly increased(P<0.05),while the CK content was not significantly different(P>0.05),while the testosterone,cortisol and T/C values in the control group were not significantly different(P>0.05),and the CK content was significantly increased(P<0.05).After intervention,compared with the control group,the concentration of testosterone and T/C in the experimental group were significantly increased,and the concentration of cortisol was significantly decreased,with statistical significance(P<0.05),while the content of CK was not significantly different(P>0.05),but the increase of CK in the control group was significantly higher than that in the experimental group(P<0.01).(2)There was no significant difference in creatine kinase content between the two groups before intervention(P>0.05).After intervention,there was no significant difference in the CK level in the experimental group(P>0.05),while the CK level in the control group was significantly increased(P<0.05).After intervention,there was no significant difference in CK content between the experimental group and the control group(P>0.05),but the change range of CK in the control group was significantly higher than that in the experimental group(P<0.01).(3)During the experiment,there was no significant difference in the positive detection rate of ten urinary indexes between the two groups(P>0.05).(4)There were no significant differences in PSQI score,sleep quality score and sleep time score between the two groups before intervention(P>0.05);After intervention,compared with before intervention,total PSQI score,sleep quality item score and sleep time item of experimental group were significantly decreased(P<0.05),while total PSQI score and sleep quality item score of control group were significantly decreased,and sleep time item score had no significant change(P>0.05).After intervention,compared with the control group,the PSQI score,sleep quality score and sleep time score of the experimental group were significantly lower than those of the control group(P<0.05).Correlation analysis between inflammatory response and functional status results:(1)There was a positive correlation between IL-18 mrna expression level and serum CK level(r=0.431,P<0.05),NLRP3 m RNA expression level and serum cortisol level(r=0.511,P<0.05).(2)There was a positive correlation between serum IL-18 and CK level(r=0.415,P<0.05),serum IL-1β and cortisol(r=0.404,P<0.05),serum IL-1β and sleep quality score(r=0.452,P<0.05).Conclusion:Ultra-low temperature whole body cryo-therapy has a certain inhibitory effect on the activation of NLRP3/caspase-1/IL-1β/IL-18 signaling pathway,which can inhibit the expression of IL-18 m RNA and reduce the levels of serum NLRP3,IL-1β and IL-18. |
