Research On Climacteric Syndrome Treated With Acupuncture Therapy Of Regulating Mentality And Supplementing Kidney

This research programme dealt extensively and deeply with climacteric syndrome treated with acupuncture therapy of regulating mentality and supplementing kidney (RMSK) from aspects of theory clinic and experiment.First, based on the long-term experience of Prof. Shi Xuemin and other experts and their understanding of "Mentality", and referred to related literature of the traditional Chinese medicine and the Taoist, the basic principle was determined that the brain store "Yuanshen (Innate mentality)" so as to control the kidney, the essence of kidney product marrow so as to support the brain, and there is a coordinative relationship between the brain and the kidney. Then, the pathogenesis, the acupuncture principle and prescription for climacteric syndrome were reelucidated, that is, deficiency of the essence of kidney is the pathogenic base, incoordination between the brain and the kidney is the pathogenic key, RMSK is the acupuncture principle, acupoint Fengfu Baihui Qihai and Shenshu is the basic prescription for acupuncture.Secondly, after treated with acupuncture therapy of RMSK, 12 cases were cured, 16 cases markedly effective and 6 cases improved in 35 cases of climacteric syndrome. The total effective rate was 97.14%. Meanwhile, the evaluated marks of symptoms were decreased significantly, the levels of serum estradiol (E2) were raised and those of serum follicular stimulating hormone (FSH) luteinizing hormone (LH) were lowered. They came to a stable condition again. Furthermore, the group treated with acupuncture therapy of RMSK was superior to the group treated with western medicine in clinical therapeutic effect improvement of the evaluated marks of symptoms, especially levels of serum hormone related to the hypothalamus-pituitary-ovary axis.Thirdly, the female rat model for climacteric was made naturally. The morphologic molecular biologic and computerized image processing techniques were adopted to observe the effect of acupuncture therapy of RMSK on the estrogen-secreting organs he neuroendocrine centre and its related factors infemale climacteric rats. The results were as follows: By cytohistochemical technique, it was showed that the content of lipoid substance in estrogen-synthesizing cells of ovary and adrenal gland were lowered in female climacteric rats. After interfered by acupuncture therapy of RMSK, the content of lipoid substance in those region were raised. These suggested that acupuncture therapy of RMSK can improve supply of raw material for synthesis of estrogen and its prosubstance in the estrogen-secreting organs. (2) By cytozymochemical technique, it was showed that the content or activity of 3p-hydroxysteroid dehyrogenase/ A isomerase (3J3-HSDH) in cells of the antral follicle and the mature corpus luteum of ovary were lowered, that of 30-HSDH in cells of the interstitial gland of ovary and cells of the zona fasciculata and zona reticularis of adrenal gland remained normal or even were increased compensatively in female climacteric rats. After interfered by acupuncture therapy of RMSK, the content or activity of 3P-HSDH in those region were increased on the former levels. These suggested that acupuncture therapy of RMSK can enhance the content or activity of the key enzyme for synthesis of estrogen in the estrogen-secreting organs. ?y technique of in situ hybridization histochemistry, it was showed that the expression of aromatase cytochrome P450(P450 AROM) mRNA in cells of the antral follicle and the mature corpus luteum of ovary were down-regulated, that of P450 AROM mRNA in cells of the interstitial gland of ovary remained normal basically in female climateric rats. After interfered by acupuncture therapy of RMSK, the expression of P450 AROM mRNA in those region, especially the antral follicle and the mature corpus luteum of ovary were increased. These suggested that acupuncture therapy of RMSK can enhance the mRNA expression of the rate-limiting enzyme for synthesis of estrogen in ovary.
