Tonifying Delay The Aging Of Ovarian Function In Old Female Rats Early Experimental Research

Objects: It is to discuss the effect and mechanism of tonifying kidney formula to delay the aging of the primary senile rat ovary and so that to testify the importance of TCM traditional classic theory " kidney governing reproduction" . The study may offer theory and experimental evidence for manufacturing efficacy new Chinese traditional medicine which can improve life quality of premenopausal women, delay the aging of ovary founction, prevent and treat relative diseases. Methods: 12-months old Wistar female rats whose oestrus elongation were set as primary senile female rat model and randomized into model control group, tonifying kidney formula in high dosage, middle dosage and low dosage group. 4-months old female rats and 18-months old female rats were selected as young control group and old control group additionally. The intervention period was 2 weeks. The blood samples were tested to detect serum E2, T, FSH and INH. The ovaries were treated by HE and Immune histochemistry method to observe growing follicles, mature follicles, corpus luteum, expression of ovary INHb, FSH , Bc1-2 and Bax. It was also treated by TUNEL to detect ovary apoptosis . The expression of right ovary INHb mRNA and pituitary FSHmRNA were measured by RT-PCR.Results: Compared with young control group, tonifying kidney formula could increase uterus index, ovary index (P<0. 05), adjust sex hormone level in blood, improve E2 and INH(P<0. 05), decrease FSH(P<0. 05); promote the growth of follicle(P<0. 05), up regulate the expression of ovary INHb and INHb mRNA(P<0. 05), weaken the expression of ovary FSH and pituitary FSHmRNA (P<0.05) In modulating H-P-O-A aspect;. In modulating ovary apoptosis aspect, tonifying kidney formula could obviously stay ovary apoptosis (P<0. 05), improve the expression of Bcl-2 in ovary (P<0. 05) and weaken the expression of Bax(P<0. 05), which could increase the ratio of Bc1-2/Bax. In free radical aspect, tonifying kidney formula could obviously increase ovary SOD(P<0. 05) and decrease ovary MDA(P<0.05).Conclusions: The results showed that tonifying kidney formula could adjust sex hormone level in blood , promote the growth of follicle, increase the expression of ovary INHb and decrease excretion of pituitary FSH, restrain ovary apoptosis. enhance the ability of recovering from free radical damage, lighten damage of aging free radical. These effects may be the mechanism of tonifying kidney formula delaying the aging of primary senile rat ovary.
