Objects:Explore the functional mechanism of herbs for "tonifying Fluid and Qi of Yang Ming" on delaying aging of the primary senile rat ovary,to testify the importance of TCM traditional classic theory "Yang Ming Meridian Declines during Wu Qi period" in premenopause with research in prophase.This research may offer a new study and therapy thought to delay the aging of ovary during premenopause, improve premenopausal women health,prevent and treat relative diseases.Methods:4~6 months old SD female rats were set to be normal control group and 10~12 months old SD female rats whose oestrus elongation were set as primary senile female rat model.Primary senile female rats were divided into 6 groups randomly as following:1.High dose Yiweitang group;2.Middle dose Yiweitang group;3.Low dose Yiweitang group;4.liuweidihuangwan group 5.Diethylstilbestrol group;6.Model control group.After 4 weeks intervention,weighing the uterus and ovaries obtained from head-off rat,using for detecting modulus of them.Getting the left ovaries and observe expression of ER,FSHR by immune histochemistry method. Then the right ovaries were treatd by hybridization histochemistry,so as to measure the expression of Bcl-2mRNA,BaxmRNA,FasmRNA,FasLmRNA,Caspase3mRNA. observeing the histologic variation and ultramicrostructure of the slices,which were made from part of the ovaries.Results:In modulating H-P-O-A aspect,compound Yiweitang which could tonify YangMing Fluid and Qi could increase uterus index,ovary index,up regulate the expression of ovary ER and FSHR.In modulating ovary apoptosis aspect, Yiweitang could improve the expression of Bcl-2mRNA in ovary and weaken the expression of Bax.It could also decrease the expression of Caspase3mRNA,FasmRNA, FasmRNA 1,and reduce the percentage of apoptosis;increasing chondriosome,smooth endoplasmic reticulum and lipid droplet of ovary granular cell,decreasing apoptotic body.Comparing with the model-controlled group,Diethylstilbestrol group and liuweidihuangwan group can promote the growth of ovarian follicle and regulate the expression of ovary ER and FSHR effectively.Comparing with Yiweitang,the function of Diethylstilbestrol group to up-regulate ER and FSHR is inferior to high-dose Yiweitang group while that of liuweidihuangwan group is no obvious difference with middle-dose Yiweitang group and is superior to low-dose Yiweitang group.Comparing with the model-controlled group,Both(Diethylstilbestrol group and liuweidihuangwan group) can restrain ovary apoptosis to some degree.Regarding the expression of Bcl-2mRNA,the diethylstilbestrol group is lower than the high-dose Yiweitang group,as for FasmRNA,FasLmRNA,Caspase3mRNA the high-dose Yiweitang group is lower.As for the expression of BaxmRNA there is no obvious disparity.The expression of Bcl-2mRNA and BaxmRNA of liuweidihuangwan group is no obviously difference with both low and middle dose Yiweitang while the expression of FasmRNA and FasLmRNA is higher than the middle-dose Yiweitang group and is no clear difference with the low-dose Yiweitang.Reguarding to the level of Caspase3mRNA liuweidihuangwan group is lower than low-dose Yiweitang group and with little difference from middle-dose Yiweitang group.conclusions:The results showed that Yiweitang,which could tonify YangMing Fluid and Qi,could change the growth internal environment of follicle,promote the growth of follicle,increase the expression of hormone receptor and restrain ovary apoptosis.These effects may be the mechanism of Yiweitang delay the aging of primary senile rat ovary.
