| The aims of the thesis are to promote the modernization and internationalization of traditional Chinese medicine(TCM)and promote the rational clinical application of traditional Chinese medicine prescription,improve the level of women’s disease diagnosis and treatment as well.Three kinds of common women diseases(mastitis,pelvic inflammation and menoxenia)were chosen as the research objects.We achieved the analysis of the compatibility rule of traditional Chinese medical formulae,the action mechanisms of formulae and the scientific interpretation the meaning of prescriptions compatibility principle about “jun-chen-zuo-shi”.The first part of the study was to conduct data mining and analysis on 3 traditional Chinese medicine(TCM)prescriptions databases(for mastitis,pelvic inflammatory disease and irregular menstruation).First of all,we collected “real clinical world”TCM prescriptions of 3 female diseases in a provincial hospital.And then we used the data mining method to analyze and excavate the frequency,the key drug combinations and new drug combinations respectively.Then the four natures,five flavours and channel tropism of each Chinese materia medica(CMM)in each database were counted.After that,based on the results obtained by data mining method,combined with the analysis of disease classification,etiology and disease syndrome TCM,the law of the use of TCM in the treatment of female diseases and the principle of TCM prescriptions are summarized.Research in the second part is to build "drug molecules-disease targets" network,"key drug combinations-disease targets/signaling pathways ” about the high frequency CMMs and key drug combinations respectively.Then the query gene functional annotation in detail and functions of signaling pathways were searched.So did the related literatures.Finally,the mechanisms of action of TCM prescriptions of 3common female diseases were summed up in this paper.The third part of the research is based on the angle of molecular pharmacology to study on the scientific meaning of compatibility principle in the classical prescription in the treatment of women’s diseases-Shenghua Decoction by building a "drug molecules-disease targets network".The “Target Database of Disease”(TDD)and the “Target Database of Shenghua Decoction”(TDSHD)were constructed.The Venn diagram was constructed to find the intersection between TDD and TDSHT(the shared target between them),and named the highly relevant target of Shenghua decoction drug molecule.Then,the diagram about relationship between network pattern of drug molecule and acting target was drawn.The key nodes with a large number of branches,that is,the key nodes of drug molecule and of target for disease,were found.Then,according to the source of the drug molecules at the key nodes,the scientificity of the“Jun-Yao”,the“Chen-Yao”,the“Zuo-Yao”and the“Shi-Yao”was verified,and the gene function of target of the disease at the key node was found to describe the mechanism of the Shenghua decoction.Based on the network pharmacology,the theory of “Jun-Chen-Zuo-Shi” and the mechanism of Shenghua decoction were scientifically interpreted from the perspective of molecular pharmacology.In conclusion,data mining,network pharmacology,genetics,molecular pharmacology method were applied to the analysis of complex system of TCM in this paper.The latest data in genetics,molecular pharmacology,TCM molecular biology are adopted as the evaluation criteria,which makes this study maintain the advantages of TCM dialectical treatment,"multiple molecules,multiple targets" and get the better convergence and integration of modern science and technology and research results.The research makes inheritance and development of TCM theory and actively explore the TCM modernization development. |
