Objective: Summarize professor Liu Tiejun’s medication rule and core prescription for stomach pain through data mining method,predict its mechanism of action by network pharmacology technology,and explain its scientificity and rationality.To provide a theoretical basis for the inheritance of the experience of experienced Chinese medicine doctors,provide an objective basis for clinical prescription and medication,and optimize the Chinese medicine treatments of stomach pain,with a view to guiding clinical practice.Methods:Research 1: Using data mining methods,analyze and sort out Professor Liu’s stomach pain cases,establish a database through the "Auxiliary Platform for Traditional Chinese Medicine Inheritance",and systematically discuss and summarize Professor Liu’s treatment experience,drug using rules and core prescriptions by using frequency statistics,association rules,cluster analysis and other methods.Research 2: On the basis of the core prescription of Research 1,it was applied to online pharmacological analysis,and the active ingredients and action targets of the core prescription were screened using STITCH and other databases;Screening of chronic gastritis related targets through Gene Cards and other databases;Adopt Cytoscape_3.7.2Software construction of compound target disease network and target protein interaction(PPI)network;The gene ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were conducted through David database and R language.Results:Study 1: 268 medical cases were included in this study,of which the disharmony between liver and stomach was the most common,followed by the stagnation of heat between liver and stomach;There were 140 drugs in total,and 20 drugs were used 80 times or more frequently.Licorice,rhubarb and cassia twig were the single drugs with the highest use frequency;The four qi medicines are mainly warm and cold;The five flavors are mainly pungent and sweet;In meridian tropism,the spleen and stomach meridians are the most common;The majority of cases were diagnosed as chronic gastritis by western medicine.Through association rule analysis,the confidence level was set to 0.9,the support level was40%,and the interview method determined that the core prescription was: "cassi-twig-gypsum-white peony-orange peel-rhubarb-licorice".Study 2: 531 core prescription active ingredients were screened,437 targets related to chronic gastritis,and 89 targets intersected with each other.The topological analysis results of the compound target disease network and PPI network were constructed,and 17 key targets such as VEGFA were screened.There are 700 GO enriched items,mainly involving positive regulation of gene expression,signal transduction and other signal pathways.134 KEGG pathways were significantly enriched,and PI3 K Akt signaling pathway was related to inflammation and immunity;MAPK signal pathway related to cell proliferation and apoptosis;The pathway of neurodegeneration related to neural regulation-multiple disease pathways,etc.Conclusion: In addition to the treatment of stomach and spleen meridians,Professor Liu Tiejun also combined the treatment of liver,heart and lung,so that Qi and can be central focus treated together.Xin and wen drugs are mostly used,which can warm and build up yang,transform qi and yin and yang,strengthen the spleen and stomach,remove dampness and cold,and dispel phlegm and ruffians;Commonly used sweet,bitter and cold medicines,take the effect of clearing heat to preserve body fluid,use cold and warm together,and balance yin and yang.When treating stomachache,we should focus on distinguishing cold and heat,deficiency and excess,and at the same time,we should not forget to dredge the qi mechanism,and we are familiar with the way of "general means no pain".In network pharmacology,core prescriptions may work together through multiple mechanisms such as inflammation and immune response,neural regulation,tumor inhibition,cell proliferation and apoptosis.The potential active ingredients and mechanism of the core prescription in treating chronic gastritis were preliminarily discussed,providing a theoretical basis for further research on the treatment of stomach pain with traditional Chinese medicine.
