Object An analytical method using high resolution inductively coupled plasma mass spectrometry(HR-ICP-MS) for rapid simultaneous determination of Na,Mg,K,Ca,Cr,Mn,Fe,Co,Ni,Cu,Zn,Se,Sr,Mo, Cd and Ba 16 trace elements in primary carcinoma of liver tissue was applied by us to investigate the effect of trace elements on etiology and pathogenesis of primary of carcinoma of liver.Methods The sample was dissolved in HNO3 and HClO4 by microwave digestion then the above 16 elements in the solution was analyzed directly by HR-ICP-MS.Most of the spectral interferences was avoided by measuring in the high resolution mode(HRM).The matrix effects due to the presence of excess Cl was evaluated.Correction for matrix effects was made by using Sc,Rh and T1 as internal standards.The optimum conditions for the determination was tested and discussed.Results 1)The detection limits of the method was in the range of 0.002~41.6μg/L,the relative standard deviation(RSD) was less than 8.35%and the recovery of the samples was in the range of 88.2%~108.2%.2)Theconcentration of Na,Cr,Mn,Ni,Co,Zn,Se,Mo and Cd in the carcinoma tissue are lower than in the health tissue.The concentrations of Na,Cr,Mn,Co,Ni,Zn,Se,Cd in the carcinomatous tissue have very remarkable differences from those in the normal tissues (P<0.01).Mo has significant differences between carcinomatous tissues and normal tissues(P<0.05).K,Ca,Sr has no significant differences between carcinomatous tissues and normal tissues(P>0.05).The concentration of Mg,Fe,Cu and Ba in the carcinoma tissue are higher than in the health tissue.The concentrations of Fe,Cu,Ba in the carcinomatous have very remarkable differences from those in the normal tissues(P<0.01).Mg has significant differences between carcinomatous tissues and normal tissues(P<0.05).Conclusions 1) The direct determination of 16 trace elements in primary carcinoma of liver tissue by HR-ICP-MS is described.The detection limits of the method are 0.002~41.6μg/mL.The recycling rate ranks from 88.2%to 108.2%.RSD is less than 8.35%.Compared with other analysis,this method is accurate,quick acquired and convenient.It is used in the determination of trace elements in primary carcinoma of liver tissue with the result is satisfactory.2)The result indicates that the change of concentration of these trace elements in primary of liver tissue is closely related to the generation of the carcinoma of liver,and this study gives clues on further research in the occurence and therapy of liver cancer.
