Objective To investigate the expression of p53protein and Ki-67antigen in humangastric carcinoma(GC),and the relation of the two parameters to biological behaviorand prognosis.Methods Tissue samples from66patients treated by gastric resection for GC from2006to2010in Hefei First People’s Hospital were used. Formalin-fixedparaffin-embedded tumors were retrospectively analysis by immunohistochemistry(MaxVisionTM2/HRP method), using monoclonal antibodies to p53and Ki-67. Theresults were correlated with clinicopathological features and prognosis. In this study66cases were successfully followed up.Results (1) Positive expression rates of p53protein and Ki-67antigen were47.10%(65/138),69.56%(96/138) respectively.(2) P53protein expression were correlated with infiltration depth, regional lymph nodemetastasis and TNM stage (P<0.05), and was not correlated with age, gender, tumorlocation, size and differentiation degree (P>0.05).(3) Ki-67expression were correlated with tumor infiltration depth(P<0.05), and wasnot correlated with age, gender, tumor location, size, differentiation degree, regionallymph node metastasis and TNM stage (P>0.05).(4) The p53protein expression in gastric carcinoma was positively correlated with theKi-67antigen expression (r=0.211, P<0.05). Combined analysis of p53protein andKi-67antigen were correlated with differentiation degree, regional lymph node metastasis and TNM stage (P<0.05), and was not correlated with tumor location, sizeand infiltration depth (P>0.05).(5) Postoperative survival time of p53protein positive expression group of patients isshorter than negative expression group (Log-rank test: χ~2=0.701, P=0.402). There is nosignificant difference of postoperative survival time between ki-67antigen positive andnegative expression group of patients (Log-rank test: χ~2=0.182, P=0.670).(6) Cox multivariate survival analysis showed that p53protein, patient age,differentiation degree were independent prognostic indicators for survival time afteroperation (P<0.05).Conclusions (1) P53protein expression was correlated with infiltration depth,regional lymph node metastasis and TNM stage.(2) Ki-67expression was correlated with tumor infiltration depth.(3) The p53protein expression in gastric carcinoma was positively correlated with theKi-67antigen expression. Combined analysis of p53protein and Ki-67antigen werecorrelated with differentiation degree, regional lymph node metastasis and TNM stage.(4) Postoperative survival time of p53protein positive expression group of patients isshorter than negative expression group. There is no significant difference ofpostoperative survival time between ki-67antigen positive and negative expressiongroup of patients.(5) p53protein, patient age, differentiation degree were independent prognosticindicators for survival time after operation.
