The present study provides a method for optimizing the extraction of phnolic acid by the combination of orthogonal design and response surface method(RSM). To establish a method for chemical pattern recognition on the quality assessment of Cimicifuga Rhizoma by hierarchical clustering analysis and principal component analysis. The determination of trace element and heavy metal element in different source of Cimicifuga Rhizoma was performed by ICP-AES. To establish a method for changing of main chemical content of Cimicifuga Rhizoma was performed by UPLC/Q-TOF-MS. We determined the plasma pharmcokinetics investigation of four kinds of phenolic acids (caffeic acid, isoferulic acid and ferulic acid) and cimifugin in rat plasma by UHPLC-MS/MS. We provided a effective and safe method for clinical reasonable medication.We determined the best three kinds of phenolic acid extraction was20times of75%ethanol, and reflux1.5h for3times by the combination of orthogonal design.,and the best total phenolic acid extraction was ultrasonic power of377.35W, extraction temperature of70℃, and ethanol concentration of58.37%.Determined three components by HPLC, the results showed three kinds of phenolic acid are not identical of different source. Cimicifuga Rhizoma in Hei Longjiang, Liaoning, Hebei province is higher than others regions. So in order to keep the curative effect from clinical use, we should be considered. Meanwhile, the chemical composition change of different souce and other products is obvious. The content of caffeic acid and ferulic acid in processed with fried and wined increased significantly. The downward trend of caffeic acid and ferlic acid in processed with honey is obvious.To establish a method for chemical pattern recognition on the quality assessment of Cimicifuga Rhizoma by hierarchical clustering analysis and principal component analysis. The study using six major composition as an index, it can more fully reflect the quality of the different souces, and more comprehensive evaluation standards than using three phenolic acid as an index.We determined the plasma pharmcokinetics investigation of four kinds of phenolic acids (caffeic acid, isoferulic acid and ferulic acid) and cimifugin in rat plasma by UHPLC-MS/MS, the result is that the cimifugin in drug-time curve has significant double peaks phenomena, and the processed products of caffeic acid in absorption elimination is faster than raw product.
