Objective: The DNA repairing mechanism of a pure urinary follicle stimulatinghormone (FSH) treatment on idiopathic oligospermia and asthenospermia had been studiedpreliminarily. By comparing the sperm density, the sperm motility, the rate of abnormalsperm, the rate of DNA fragmentation and the malonaldehyde (MDA) level before andafter treatment with FSH injection, we demonstrated that FSH can improve the malefertility, and preliminarily revealed the DNA repairing mechanism of a pure urinary folliclestimulating hormone (FSH) treatment on idiopathic oligospermia and asthenospermia.Methods:The148patients for male infertility with idiopathic oligospermia andasthenospermia from Shandong university of Chinese medicine second affiliated hospitalreproductive center in November2012to August2013were selected. All the patients weredeparted into3groups randomly:50were in Shengjing san group,49were in FSH group,and49were in placebo group. The patients from Shengjing san group took Shengjing san,one day with2.3grams. The patients from FSH group took FSH, once every2days, andintramuscular injection. The patients from placebo group took placebo, one day with2.3grams. By comparing the above indexes of the patients before and after treatment everymonth, especially the rate of DNA fragmentation, we preliminarily revealed themechanism of action of FSH. Results:1. Comparison of the total clinical efficacy amongeach group: FSH group and Shengjing san group have a better clinical efficacy thanplacebo group on the basis of statistics (P<0.05). There is no statistical diffenencebetween FSH group and Shengjing san group (P>0.05).2. Comparison of the spermdensity among each group: The sperm density after treatment of FSH group and Shengjingsan group is higher than that befor treatment on the basis of statistics (P<0.05) respectively, and higher than that of placebo group (P<0.05). After treatment, the spermdensity of FSH group is more than Shengjing san group on the basis of statistics (P<0.05).3. Comparison of the sperm motility among each group: The sperm motility PR andPR+NP after treatment of FSH group and Shengjing san group is higher than that befortreatment on the basis of statistics (P<0.05) respectively, and higher than that of placebogroup (P<0.05). However, there is no statistical diffenence between FSH group andShengjing san group (P>0.05).4. Comparison of the rate of abnormal sperm among eachgroup: The rate of abnormal sperm after treatment of FSH group is less than that befortreatment on the basis of statistics (P<0.05), and less than that of Shengjing san group andplacebo group (P<0.05). However, there is no statistical diffenence between Shengjingsan group and placebo group (P>0.05).5. Comparison of the rate of DNA fragmentationamong each group: The rate of DNA fragmentation after treatment of FSH group andShengjing san group is less than that befor treatment on the basis of statistics (P<0.05)respectively, and less than that of placebo group (P<0.05). However, there is no statisticaldiffenence between FSH group and Shengjing san group (P>0.05).6. Comparison of theMDA level among each group: The MDA level after treatment of FSH group andShengjing san group is less than that befor treatment on the basis of statistics (P<0.05)respectively, and less than that of placebo group (P<0.05). However, there is no statisticaldiffenence between FSH group and Shengjing san group (P>0.05). Conclusion: Thisstudy shows that there exists a certain correlation between the sperm motility and the rateof DNA fragmentation. FSH can improve the sperm motility and decrease the the rate ofDNA fragmentation through decreasing the MDA level. FSH can improve the spermquality (including the sperm density, the sperm motility PR and PR+NP, and the rate ofDNA fragmentation), and increase the male fertility. In addition, there exists positivecorrelation between the rate of DNA fragmentation and MDA level.
