Objective: To investigate the effect of lysine specific demethylase 1(LSD1),O6-methylguanine-DNA methyltransferase(MGMT),nuclear associated antigen Ki-67 on the grade and prognosis of high-grade glioma.Methods: Sixty-five patients with high-grade glioma specimens were selected.Immunohistochemistry(SP method)was used to detect the expression of LSD1,MGMT and Ki67 protein in pathological specimens.The long-term follow-up was used to evaluate the therapeutic effect and to analyze the relationship between the three markers and pathological grade,progression-free survival,overall survival,and analyze whether the three have correlation or not.Results: The overall positive rates of LSD1,MGMT and Ki67 in 70 high-grade glioma specimens were 70.8%(46/65),60.0%(39/65),and 100%(65/65).There was no significant difference in the expression of LSD1 and MGMT in grade ? and ?gliomas(P=0.420),(P=0.908).The expression of Ki67 in grade ? gliomas was higher than that in grade ? gliomas,and the difference was statistically significant(P=0.037).Log-rank analysis showed that the positive expressions of LSD1,MGMT and Ki67 were negatively correlated with PFS of high-grade glioma(P=0.007,0.035,0.002),and negatively correlated with OS(P=0.008,0.01,0.003).Patients with grade ? grade were more likely to have recurrence progression than grade ? patients(P=0.010),and overall survival was shorter(P=0.046).Multivariate analysis found that Ki67 and LSD1 were independent prognostic factors for high-grade glioma.The HR of Ki67 and PFS is 1.703,and the HR of OS is 1.778.The HR of LSD1 and PFS is 1.361,and the HR of OS is1.406.Spearman rank correlation analysis revealed a positive correlation between MGMT and LSD1 expression in high-grade glioma.Conclusion: MGMT is a relative factor of prognosis for high-grade glioma.Ki67 and LSD1 can be used as independent predictorsof high-grade glioma prognosis.MGMT was positively correlated with LSD1 expression in high-grade glioma.
