Applying simple random sampling in typical colony method,63 Hu Sheep and 65 Tong Sheep were sampled in their central area of habitat Huzhou city Zhejiang province and Baishui county Shanxi province respectively.Each individuals had respective bloodline.The body measurements were measured and the phenotype frequency of relative appearance characters were observed.Starch gel electrophoresis was used to detect the variations of 14 blood enzyme and other protein structural loci,and the reliability and accuracy of every estimated frequencies value were evaluated.It also detected the polymorphism of 7 "microsatellite" DNA site which distributed on 6 different chromosome.At the same time,49 Yangtse River Delta White Goats were sampled as contrast population from the suburb of Yangzhou city Jiangsu province by the simple random sampling method.The results were as follows: 1. The content of gene library were displayed by the appearance characters , polymorphism of blood enzyme and other proteim polymorphism of "microsatellite" DNA and other genetic markers. It reflected the different genetic construction and breed features of Hu Sheep and Tong Sheep.2.Among the 14 detected blood enzyme and other protein structural loci,Hu Sheep population had 12 polymorphism loci,its dominant gene were P0s,TfD, AlpB-,Ary-EsEs-, LapB, Hb-βB, X-px CAS, CatC, LyA, Keh.Tong Sheep population had11 polymorphism loci,its dominant gene were TfC, AlpB-, Ary- EsEs+, LapB, Hb-β B,X-px, CAS, MDHs, CatC, LyA, Keh.3.The average heterozygosity of blood enzyme and other protein structural loci were0.3018 and 0.3253 in Hu Sheep population and Tong Sheep population respectively.Itindicated that Hu Sheep population had lower genetic variations than Tong Sheeppopulation.4. Seven "microsatellite" site had high polymorphism in Hu Sheep population and Tong Sheep population.Their average heterozygosity were 0.9096 and 0.9177.Their average polymorphism information content were 0.9024 and 0.9116. It also indicated that Hu Sheep population had lower genetic variations than Tong Sheep population.5. After collecting the same data about 21 sheep populations of China and external ,it extablished and composed the matrix of fuzzy similarity relation describing the genetic similarities of different populations basing on the alleles frequencies of 11 blood enzyme and other protein structural loci including Hu Sheep population , Tong Sheep population and Yangtse River Delta White Goat population. 24 populations were also clustered according to their phylogenetic relationship.This study proved that Hu Sheep population was closer to Tong Sheep population in their phylogenetic relationship,and they were relatively closer to Mongolia populations,and perhaps they shared the same origin in nearer times.This result obtained was shown to conform to the historical data.
