In order to illustrate the anti-inflammation and curative effect mechanisms of the main compound recipe of traditional chinese medicine for edema disease of swine,we investigated the effect of SLT-Ⅱe on secretion function and the expression of CAMs in rat intestinal microvascular Endothelial cells(RIMEC) in vitro,and the effect of the main compounds recipe of traditional chinese medicine(pachymic acid, astragaloside IV, astragalus polysaccharin, emodin, rhein, hyperin, quercetin) on the abnormality of secretion function and the expression of CAMs in RIMECs induced by SLT-Ⅱe.1.We established animal model of edema disease of swine and filtrated the best recipe of traditional chinese medicine for edema disease of swine.2. We established cell model of edema disease of swine for 1μg/mL SLT-Ⅱe stimulating RIMECs and increased the abnormality secretion of NO, ET-1,PGI2, TXA2, P-selectin,sICAM-1 production in RIMECs.3.We focused the effect of pachymic acid on secretion function and the expression of CAMs in RIMECs by SLT-Ⅱe with ELISA, deoxidization enzyme and in situ hybridization(ISH). The results showed that pachymic acid had inhibited the secretion of SLT-Ⅱe induces NO,ET-1, TXA2,sICAM-1 and the expression of iNOS mRNA,ecNOS mRNA,ET-1 mRNA,ICAM-1 mRNA production in RIMECs.4. We observed the effect of astragaloside IV and astragalus polysaccharin on secretion function and the expression of CAMs in RIMECs by SLT-Ⅱe with ELISA, deoxidization enzyme and ISH. The results showed that astragaloside IV had inhibited the secretion of SLT-Ⅱe induces NO,ET-1, sICAM-1 and the expression of iNOS mRNA,ET-1 mRNA,ICAM-1 mRNA production in RIMECs. The results also showed that astragalus polysaccharin had inhibited the secretion of SLT-Ⅱe induces NO,ET-1,TXA2, sICAM-1 production in RIMECs.5. We investigated the effect of emodin and rhein on secretion function and the expression of CAMs in RIMECs by SLT-Ⅱe with ELISA, deoxidization enzyme and ISH. The results showed that emodin had inhibited the secretion of SLT-Ⅱe induces NO,ET-1, PGI2,P-selectin,sICAM-1 and the expression of iNOS mRNA,ET-1 mRNA,ICAM-1 mRNA and increased the expression of AQP1 production in RIMECs. The results also showed that rhein had inhibited the secretion of SLT-Ⅱe induces NO,ET-1, P-selectin,sICAM-1 production in RIMECs.6. We discovered the effect of hyperin and quercetin on secretion function and the expression of CAMs in RIMECs by SLT-Ⅱe with ELISA, deoxidization enzyme and ISH. The results showed that hyperin had inhibited the secretion of SLT-Ⅱe induces NO,ET-1, TXA2,sICAM-1 and the expression of iNOS mRNA, ET-1 mRNA,ICAM-1 mRNA production in RIMECs. The results also showed that quercetin had inhibited the secretion of SLT-Ⅱe induces NO, ET-1, PGI2, TXA2, sICAM-1 production in RIMECs.Our results indicate that the anti-inflammation and curative effect mechanisms of the main compound recipe of traditional chinese medicine for edema disease of swine may inhibited the abnormality secretion of NO,ET-1,PGI2,TXA2,P-selectin, ICAM-1 production in RIMECs, adjusted the secretion of AQP1 and ameliorated microcirculation.
