Hubei Maidong is the main source of Radix Liriopes recorded in the "People's Republic of China Pharmacopoeia"(1995,2000 and 2005 edition).Liriope spicata var.prolifera is the special species in Hubei Province.as the As the chief variety of ophiopogon japonicas commercial medicinal materials,planting area of Liriope spicata var. prolifera has reached more than 5 acres,with an annual output near 10,000 tons accounting for 50%of the National Medicinal Ophiopogon output.Liriope spicata var.prolifera has a long medicinal history.Suizhou Ophiopogon which was recorded by "Bencao Tujing " and " Zhenglei Bencao " early in the Song Dynasty,according to their drug plan research,is the same as Liriope spicata var.prolifera used today.However,cultivation,yield and quality of Hubei Ophiopogon is instability in 40 years.So it is difficult to guarantee the quality of medicines.It is necessary to standardize plant study to improve its productivity and quality.The project of"Liriope spicata var.prolifera standardization demonstration bases and plant construction" was confirmed established by by the Science and Technology Hall of Hubei in 2001.Study of standard planting Liriope spicata var.prolifera obtained much supply of funds such as the Ministry of Science and Technology and the Science and Technology Hall of Hubei.Studying team made technical research about Liriope spicata var.prolifera on group germplasm resources in Liriope spicata var.prolifera investigate,germplasm identification,species improvement,environmental quality assessment, nutrition physiology,harmless plant technology,product quality standards,etc.Standard planting According to GAP's request has provided a strong technical support for developing Liriope spicata var. prolifera production.In the study of standard planting,what have been mainly considered were the growth characteristics of Liriope spicata var. prolifera of Liriope spicata var.prolifera history and scale,the ecological environment of origin,sources and identification of varieties,germplasm sources,the distribution of wild resources,growth habit,seed breeding, the suitable harvest period,the situation of integrated pest management, quality control and plant and flow control of key technologies,the origin of ecological environment Detect report(including soil,irrigation water, atmospheric environment),quality standards,sampling methods, procedures,management protocols,technical standards,and job responsibilities.A complete production and quality system of cultivation, planting and primary processing has been formed by researching and the quality and output of Liriope spicata var.prolifera has been enhanced through popularization and application in producing.polysaccharides and saponins are the active components of Liriope spicata var.prolifera contained active ingredients.However,only polysaccharides or only Saponins was extracted from Liriope spicata var.prolifera and often resulted in another major category of the active ingredients of the waste is not conducive to the comprehensive utilization of Liriope spicata var.prolifera.For Maidong extraction,which is also a frequent occurrence.The use of the subject by Macroporous Adsorption Resin Liriope spicata var.prolifera synchronous extraction of total saponins and total polysaccharide,there is in favor of Liriope spicata var.prolifera total polysaccharides and total saponins utilization, at the same time,as well as the comprehensive utilization of Maidong reference.Reported in the literature,the Liriope spicata var.prolifera total polysaccharides extraction and purification process is more complicated.For example,someone will Liriope spicata var.proiifera after defatted with petroleum ether,followed by ethanol,water extract, water extract combined,concentrated,concentrated liquid followed by Savage Removing protein,activated carbon decolorization,and then ethanol precipitation,followed by sediments with 95%ethanol, anhydrous ethanol,ethyl ether,acetone washing,drying,a crude polysaccharide(a rate of 7%).Some use of 0.9%NaCl extract Liriope spicata var.prolifera,using chloroform - n-butanol in addition to proteins, dialysis in addition to small molecule impurities,concentrated, concentrated liquid ethanol precipitation,followed by sediment with anhydrous ethanol,acetone,ethyl ether washing,drying,total polysaccharides.More than two ways to extract,refined many steps to operate more complicated,therefore more active ingredients loss, product yield is low.Liriope spicata var.prolifera have to be followed by using ethanol,water extract,water extract of the merger,after microfiltration,ultrafiltration using spiral roll system,ultrafiltration, concentration,drying,different molecular weight distribution of the polysaccharide.However,ultrafiltration is only the size of the basis of molecular weight extract of traditional Chinese medicine classification of components in the application of traditional Chinese medicine production is still small.Mainly because the active ingredients of traditional Chinese medicine complex,easily give rise to membrane contamination,coupled with the high cost of membranes and other reasons,so that the technology has so far produced in Chinese medicine should not be applied.In this paper,macroporous resin extraction method synchronization of Liriope spicata var.prolifera total saponins and total polysaccharide,these small,simple process,easy to operate and easy to industrialization,is worth popularizing.Diabetes is a common metabolic disease,its incidence has increased year by year.Maidong was proven hypoglycemic effect,the subject through the experimental results show that total polysaccharides Maidong Hubei,Hubei Ophiopogon total saponins, Liriope spicata var.prolifera total extract and the positive drug,like Delta,Culture and Sport compared with the model group were significantly differences illustrate Liriope spicata var.prolifera on alloxan-induced diabetic rats had significant hypoglycemic effect,for the treatment of diabetes Liriope spicata var.prolifera Clinical applications and new drug development to provide an experimental basis.Although experimental results show that a variety of Liriope spicata var.prolifera extract the role of less than positive drug Meikang Tatsu hypoglycemic effect,but found that the total extract of Ophiopogon japonicus Hubei and Liriope spicata var.prolifera total polysaccharides hypoglycemic effect the first 14 days than the first 7 days,some better, efficacy with the passage of time has gradually increased trend.We speculated that a longer period of time the sustainable use of Liriope spicata var.prolifera hypoglycemic effect probably better,this requires further experimental confirmation. According to this research,we calculated that Hubei Maidong polysaccharide crude extraction of total yield was 17.45%,while Hubei Ophiopogon total saponins in crude extraction yield is only 0.75%,therefore,a separate extraction of saponins for reducing glucose treatment is not practical,extracting polysaccharide is better.Hubei Maidong total extraction(water extraction) of the amount is 5ml/kg/d, equivalent to the original medicinal 5g/kg/d.based on calculations,the total content of polysaccharides and total saponins were seperately 0.8725g and 0.0375g,which is greater than the total amount of 0.4g/kg/d,but the experimental results have shown that the reducing glucose effect is lower than Hubei Maidong total polysaccharides,probably because Hubei Maidong contained small molecule of glucose.Therefore,when Hubei Maidong is used for reducing glucose treatment,it is necessary to appropriately extract impurities,there can be a better effect.In this paper,we established the use of ISSR molecular marker technology category Maidong study the genetic diversity of medicinal method for the first time,we have screened out of nine ISSR primers,15 samples of genetic characteristics and phylogenetic relationship analysis.From the correlation and cluster analysis results,Hubei Maidong,Chuanmai winter and broad-leaved Ophiopogon japonicus, tenuissimum Maidong major groups,such as classification consistent with the traditional form.Compared with RAPD,ISSR molecular marker technology has the advantages of good reproducibility and stability,which is a fast and reliable DNA fingerprinting technology,and has been widely used in wheat,potatoes,oranges,apples and other crops to identify the species, as well as schisandrin,Crocus sativus,Magnolia,Polygonatum odoratum,Bupleurum Chinese varieties,such as the relationship between genetics and the identification of a preliminary study.It is the first time that this issue is applied to study medicine category Maidong through 9 of the amplification primers and completed between different medicines Maidong identification of molecular markers,which indicate the results of ISSR molecular marker technique can be used for genuine medicines Maidong category and the identification of related species.
