Paclobutrazol is active member of the triazole family, which is widely used as plant growth regulator. It reduces the quantity of gibberellins biosynthesis in the plants, so it can inhibit the elongation of plant stems and let the stems get sturdy, promote branching, tiller, root, flower and fruit, strengthen drought resistance and cold hardiness, dwarf plants, increase yield, also enhance pest resistance,Liriope spicata var.prolifera is famous-region drug of Hubei province, it is widely planted in Oumiao town Xiangfan city Hubei province,its annual yield is more than sixty percent of the national total yield. The local farmers have the used the paclobutrazol habit. The application of paclobutrazol in Liriope spicata var.prolifera has not been reported. In order to further regulate the planting of Liriope spicata var.prolifera,the experiment studies on different quantity and time for the local farmers provide the basis for reasonable use of paclobutrazol.Liriope spicata var.prolifera was used as the experimental subject, which is plant in Oumiao town Xiangfan city Hubei provine. Effect of different quantity and time of paclobutrazol on yields and quality of Liriope spicata var.prolifera and the residues in herbs and the soil. The application of paclobutrazol quantity is 0g/m~2, 0.42g/m~2, 0.84g/m~2, 1.26g/m~2, 1.68g/m~2,the application of paclobutrazol time is July6,2008,September19,2008, November10,2008.Choosing a non-continuous crop -ping land, the land was identified into three big field blocks by the three application of paclobutrazol time, then let each big field block divide into fifteen small blocks, the area of each small block is 2.8 square meters(140cm×200cm), the five doses of paclobutrazol were used in the fifteen small blocks. Each dose of paclobutrazol was used by repeating three small blocks. The span is 30 cm between small block. The distance between plants is 10cm,the distance between rows is 15cm. Taking paclobutrazol powders and adding equivalence phosphate fertilizer, then furrow application. Liriope spicata var.prolifera were planted in the field blocks by the management including watering,fertilization and anti-pest measures. When the Liriope spicata var.prolifera is mature, ripe root tubers were collected and prepared March 27,2009.The content of polysaccharide in the Liriope spicata var.prolifera was determined by method of phenol-sulfuric acid,quantitative determination of Liriopesides B was tried by using the detector ELSD in combination with HPLC,the chromatographic condition was as follows:column:Agilent TC-C18(4.6×250μm 5μm); The temperature of the column:40℃; Mobile phase:acetonitrile-methan-ol- water(65:5:35); Mobile phase rate:1mL/min. The parameters of detector ELSD were:Drift tube temperature:93.2℃. The content of the two kinds of active compounds were used in quality evaluation.The paclobutrazol residue was measured by gas chromatography. The chromatographic condition in Liriope spicata var.prolifera:Capillary column:Agilent HP-5 (30.0m×320μm×0.25μm); Carrier gas:N2; Mobile phase rate:5.5mL/min; The temperature of sample injector 280℃; The temperature of detector:280℃; Splitless injection; The quantity of sample injection:1μL; Air rate:450mL/min; Hydrogen gas rate:40mL/min; Make-up gas rate:45mL/min; The temperature programming of capillary column:initial temperature 150℃, holding 1min, then heating-rate 10℃/min,temperature to 180℃, holding 2min,then heating-rate 20℃/min,temperature to 250℃, holding 4min. The chromatographic condition in the soil was as follows:Capillary column:Agilent HP-5 (30.0m×320μm×0.25μm); Carrier gas:N2; Mobile phase rate:5.0mL/min; The temperature of sample injector 280℃; The temperature of detector:280℃; Splitless injection; The quantity of sample injection: 1μL; Air rate:450mL/min; Hydrogen gas rate:40mL/min; Make-up gas rate:45mL/min; The temperature programming of capillary column:initial temperature100℃, holding 1min, then heating-rate 20℃/min, temperature to 250℃, holding 5min. Additionally, we determined the output of each quadrat and calculated output of each acreage according to the area. Data annlysis used SPSS15.0 One-Way ANOVA method was used to compare the different quantity and time of paclobutrazol on yields and quality of Liriope spicata var.prolifera and the residues in herbs and the soil. The results ale as follows:1. Comparisons among the different quantity of paclobutrazol for Liriope spicata var.prolifera(1)the yield: 1.68g/m~2 >0.84g/m~2 >1.26g/m~2 >0.42g/m~2≈0g/m~2;(2)the content of polysaccharide: 1.68g/m~2 >0.84g/m~2 >1.26g/m~2 >0.42g/m~2 >0g/m~2;(3)the content of Liriopesides B: 1.68g/m~2 >0.84g/m~2>1.26g/m~2>0g/m~2> 0.42g/m~2.2. Comparisons among the different time of paclobutrazol for Liriope spicata var.prolifera(1)the yield: November 10 > September 19 > July 6;(2)the content of polysaccharide: September 19> November 10 > July 6;(3)the content of Liriopesides B: July 6> November 10> September 19.3. Comparisons among the different quantity of paclobutrazol for Liriope spicata var.prolifera(1)the paclobutrazol residue in Liriope spicata var.prolifera: 0.42g/m~2>0g/m~2> 1.68g/m~2>0.84g/m~2>1.26g/m~2;(2)the paclobutrazol residue in the soil: 1.68g/m~2>0g/m~2>0.42g/m~2>1.26g/m~2 >0.84g/m~2.4. Comparisons among the different time of paclobutrazol for Liriope spicata var.prolifera(1)the paclobutrazol residue in Liriope spicata var.prolifera: July 6>November 10>September 19;(2)the paclobutrazol residue in the soil: November 10> September 19> July 6.Conclusion: Reasonable quantity of paclobutrazol in Liriope spicata var.prolifera is 0.84g/m~2, the best time of paclobutrazol in Liriope spicata var.prolifera is September late.
